Increased Migration of Monocytes in Essential Hypertension Is Associated with Increased Transient Receptor Potential Channel Canonical Type 3 Channels

Increased transient receptor potential canonical type 3 (TRPC3) channels have been observed in patients with essential hypertension. In the present study we tested the hypothesis that increased monocyte migration is associated with increased TRPC3 expression. Monocyte migration assay was performed in a microchemotaxis chamber using chemoattractants formylated peptide Met-Leu-Phe (fMLP) and tumor necrosis factor-alpha (TNF-alpha). Proteins were identified by immunoblotting and quantitative in-cell Western assay. The effects of TRP channel-inhibitor 2-aminoethoxydiphenylborane (2-APB) and small interfering RNA knockdown of TRPC3 were investigated. We observed an increased fMLP-induced migration of monocytes from hypertensive patients compared with normotensive control subjects (246+/-14% vs 151+/-10%). The TNF-alpha-induced migration of monocytes in patients with essential hypertension was also significantly increased compared to normotensive control subjects (221+/-20% vs 138+/-18%). In the presence of 2-APB or after siRNA knockdown of TRPC3 the fMLP-induced monocyte migration was significantly blocked. The fMLP-induced changes of cytosolic calcium were significantly increased in monocytes from hypertensive patients compared to normotensive control subjects. The fMLP-induced monocyte migration was significantly reduced in the presence of inhibitors of tyrosine kinase and phosphoinositide 3-kinase. We conclude that increased monocyte migration in patients with essential hypertension is associated with increased TRPC3 channels.

Involvement of Inflammasome Components in Kidney Disease

Nrf2 Activation in Chronic Kidney Disease: Promises and Pitfalls

Pretransplant characteristics of kidney transplant recipients that predict posttransplant outcome

Summary: The characterization of potential kidney transplant recipients using novel biomarkers can lead to improved outcomes after transplantation. This mini-review focuses on the current understanding of pretransplant recipients' characteristics, biomarkers, and immunology. Clinical characteristics and immunological risk assessment, such as histocompatibility leukocyte antigens (HLAs) and non-HLA antibodies, have been used to predict recipient and allograft survival. Recent studies have shown that pretransplant markers, such as plasma endotrophin levels and non-synonymous single-nucleotide polymorphism mismatch, are associated with posttransplant complications and allograft loss. Integrating clinical characteristics and novel markers is important for making better diagnostic decisions tailored to individual patients.

Head-to-Head Comparison of Oxidative Stress Biomarkers for All-Cause Mortality in Hemodialysis Patients

Successful low-dose immunotherapy after kidney transplantation in a 10-year-old girl with Schimke immuno-osseous dysplasia

Endotrophin Levels Are Associated with Allograft Outcomes in Kidney Transplant Recipients

Summary: This study evaluated endotrophin (ETP) as a prognostic marker in kidney transplant recipients. The measurement of collagen type VI formation could aid in predicting graft function in kidney transplant recipients.

Nrf2 Protein Serum Concentration in Human CKD Shows a Biphasic Behavior

Summary: Oxidative stress plays a role in the progression of CKD and related mortality. Nrf2, an important regulator of cellular redox status, is being evaluated for its therapeutic potential in CKD. In this study, Nrf2 protein concentrations were analyzed in CKD patients at different stages and in healthy subjects. The results showed that Nrf2 protein was upregulated in mild to moderate kidney function impairment, but reduced in severe impairment. These findings suggest the need to explore the efficacy of Nrf2 therapies in specific populations of CKD patients.

Lower levels of FOXP3 are associated with prolonged inflammatory responses in kidney transplant recipients

Metabolic regulation of forkhead box P3 alternative splicing isoforms and their impact on health and disease

Summary: FOXP3 is a crucial protein for the development and function of T cells, and its splicing isoforms have different effects on T cell differentiation and suppressive function. The balance of FOXP3 isoforms is related to various diseases, and the regulation of energy metabolism and signaling pathways can affect FOXP3 transcription and splicing. Therefore, targeting FOXP3 splicing isoforms in Tregs may provide potential approaches for the treatment of certain diseases.
