Summary: Our study demonstrates that the PHA-based SNA (PHAAD) can significantly enhance transdermal drug delivery, improve cellular uptake, down-regulate TIMP-1 expression, and enhance the cytotoxicity of DOX. Additionally, PHAAD nanoparticles facilitate hypertrophic scar cell apoptosis, reduce scar burden, and progression in a xenografted mouse model without adverse effects.

Summary: Overcoming cisplatin-based drug resistance in lung cancer remains a challenge globally. Recent studies have shown that Rab GTPases play a role in multiple aspects of tumor progression, including drug resistance. In this study, a nanosystem was developed using Rab26 siRNA-loaded nanoparticles, which effectively transfected cisplatin-resistant lung cancer cells and induced apoptosis while inhibiting autophagy disruption. The combination therapy of siRab26 knockdown with cisplatin improved antitumor therapy in vitro and in vivo. These findings suggest that siRNP is an effective platform for lung cancer therapy in cases of drug resistance.

Summary: Lipid-based nanocarriers have shown great potential in delivering genetic material, but PEGylation can have adverse effects on cellular uptake and endosomal escape. This study proposes amphiphilic poly(N-methyl-N-vinylacetamide) (PNMVA) derivatives as alternatives to lipid-PEG for siRNA delivery. DSPE-PNMVA(24) demonstrates comparable stealth properties to DSPE-PEG, but without adverse impact on cellular uptake and endosomal escape. In vivo studies with mice show that DSPE-PNMVA(24) lipoplexes have good stealth properties, extended circulation time, reduced immunological reaction, and no systemic pro-inflammatory response.

Summary: Rod-shaped nanoparticles made of C-S-B triblock polypeptides show efficient delivery of siRNA into cells, leading to gene knockdown and apoptosis. However, their lower effectiveness in inducing cell death compared to cationic lipid-based formulations is attributed to high lysosome co-localization, which requires future optimization. Overall, non-toxic C-S-B nanoparticles warrant further development as a new siRNA delivery system for cancer therapy.

Summary: Anaplastic thyroid carcinoma (ATC) is a highly lethal subtype of thyroid cancer that does not respond well to traditional treatments. This study developed specialized gold nanoparticles (AuNPs) for photothermal therapy (PTT) targeted specifically at ATC. The functionalized holo-transferrin AuNPs showed the best efficacy for PTT against ATC and demonstrated good safety and biodistribution when administered in vivo. These results suggest that AuNPs-mediated-PTT could be a promising treatment for ATC in the future.

Summary: A novel bone-affine squalene-based nanomedicine was developed by conjugating a hydroxybisphosphonate moiety to the squalene chain and inserting it into squalenoyl-gemcitabine nanoparticles. The presence of squalenyl-hydroxybisphosphonate enhanced nanoparticles' binding affinity for hydroxyapatite in bones and preserved their anticancer activity in vitro. Further in vivo studies are needed to explore the potential of these nanomedicines for treating bone sarcomas.

Summary: Biocompatible nanoscale iron carboxylate metal-organic frameworks (nanoMOFs) show potential in delivering therapeutic molecules efficiently, but their efficacy is hindered by the sequestration of loaded drugs in lysosomes, preventing them from reaching the nucleus, the subcellular target of the drug. Further optimization is needed to enhance successful drug delivery.
