γH2AX Assay as DNA Damage Biomarker for Human Population Studies: Defining Experimental Conditions

H2AX histone phosphorylation represents an early event in the cellular response against DNA double-strand breaks (DSBs), and plays a central role in sensing and repairing DNA damage. Therefore, the analysis of H2AX phosphorylated (gamma H2AX) may be possibly used as biomarker of genotoxicity and genomic instability with a number of applications in human epidemiology. However, the lack of an experimental standard leads to a wide heterogeneity in the results obtained and their interpretation, affecting the reliability of the assay. To address the most critical issues limiting the use of the gamma H2AX assay in human population studies, a flow cytometry analysis was performed to establish differences in gamma H2AX levels between fresh or cryopreserved peripheral blood lymphocytes, and to assess the influence of phytohemagglutinin (PHA) stimulation. To this purpose, cells were treated with 4 known genotoxic chemicals with different mechanisms of DSB induction, ie, bleomycin, methyl methanesulfonate, camptothecin, and actinomycin. According to our results, both unstimulated and stimulated fresh lymphocytes can be efficiently employed to evaluate gamma H2AX levels, but the sensitivity of the assay is depending upon the kind of damage observed. On the other hand, cryopreserved lymphocytes require PHA stimulation since unstimulated cells showed too high basal damage. Consequently, the protocol conditions will depend on the expected mechanism of production of DSB and the characteristics of the study design (sample collection and storage conditions, type of epidemiological study). Further studies are required to standardize the protocol of gamma H2AX assay to be employed as biomarker of genotoxicity or genomic instability in human population studies.

Summary: This study analyzed the association between TTV viremia and physical frailty and cognitive impairment. The results showed that TTV viremia was associated with physical frailty and cognitive impairment. However, the association varied in different study samples.

This review focuses on the toxicity assessment of inhaled (nano)particulate materials and provides an overview of available human respiratory models and exposure systems for in vitro testing, as well as their advantages, limitations, and current research.

Summary: Measurement of DNA migration in the comet assay can be done through image analysis or visual scoring, with the latter accounting for a significant portion of the published results. This study examines the variability in visual scoring of comets among different investigators and suggests the use of three reference sets for scoring. The results show both inter-investigator and intra-investigator variation in scoring, indicating a need for further standardization, but also demonstrate that visual scoring remains a reliable method for analyzing DNA migration in comets.

Summary: The study finds that potassium bromate is a suitable positive control for the Fpg-modified comet assay, which is widely used to measure oxidatively generated damage to DNA. Although all laboratories obtained positive concentration-response relationships, there was variation in the levels of Fpg-sensitive sites between different laboratories. However, the consistency between the measurements of %Tail DNA values and visual scores of comet images done in the central laboratory suggests a potential source of inter-laboratory variation.

Multiparametric in vitro genotoxicity assessment of different variants of amorphous silica nanomaterials in rat alveolar epithelial cells

Summary: This study assessed the cytotoxicity and genotoxicity of different-sized and surface-modified amorphous silica nanomaterials on rat alveolar epithelial cells. The results showed that variants with larger surface areas had higher cytotoxic potential, SiO2_40 induced significant DNA damage, and all tested variants increased gamma-H2AX levels. This study supports the use of multiparametric approaches to improve understanding of nanomaterial mechanisms and hazard prediction.

Effect of Air Pollution on the Basal DNA Damage of Mother-Newborn Couples of México City

Summary: DNA damage in newborn children's cord blood cells from Mexico City is correlated with the DNA damage in their mothers, as well as the environmental exposure and complications during pregnancy. Mothers living in the southern part of the city have double DNA damage compared to those living in the northern part. This study highlights the importance of antioxidants during pregnancy and the role of the placental barrier in protecting newborns from DNA-damaging effects of pollution.

Effects of Zinc Oxide Nanoparticle Exposure on Human Glial Cells and Zebrafish Embryos

Summary: The aim of this study was to investigate the effects of zinc oxide nanoparticles (ZnO NPs) on human glial cells and zebrafish embryo development, as well as to explore the role of released Zn2+ ions in these effects. The results showed that exposure to ZnO NPs caused morphological alterations and a significant decrease in cell viability. Zebrafish embryos also exhibited a decrease in hatching rate. The study further found that these effects were partly attributed to the released Zn2+ ions.
