Summary: A rhodium-catalyzed hydrothiolation reaction of terminal allenes with thioacids has been developed for the synthesis of chiral branched allylic thioesters. The reaction exhibits excellent regio- and enantioselectivity and is applicable to the diastereoselective synthesis of thioesters from Fmoc-protected aminothioacids.

Summary: A CuI-catalyzed C-N coupling reaction between 3-bromoDMAP and L-prolinamides was carried out under 80°C for 12-16 hours, where the structure of prolinamide exerted an accelerating effect on the Ullmann type reaction. This reaction was utilized for the construction of chiral 3-amino DMAP catalysts. Moreover, enantioenriched DMAP analogue C8 was applied in an asymmetric Black rearrangement of 2-benzofuranylcarbonates, leading to the formation of 3,3-disubstituted benzofuran-2-ones with up to 96% yield and 97% ee.

Summary: The search for novel, physiologically active imidazoles is an exciting research topic for medicinal chemists. Imidazole, a five-membered aromatic heterocycle, is present in both natural and synthesized compounds. Current anticancer drugs have limitations such as toxicity and limited efficacy, highlighting the need for new chemotherapeutic agents. Imidazole development as a reliable and safer alternative to anticancer treatment is gaining attention, with potential targets including tubulin/microtubule polymerization inhibition and changes in DNA structure and function, VEGF, topoisomerase, kinases, histone deacetylases, and other proteins involved in gene expression.

Summary: Temporin-PF (TPF) was identified as a peptide effective against Gram-positive bacteria but not Gram-negative bacteria. MD simulation showed that the peptides could form oligomers, but substitution of Trp/D-Trp enhanced inter-peptide interaction and eliminated overall biological activities.

Summary: The discovery of new rotihibin analogues in Streptomyces scabies, a major pathogen causing common scab disease, provides insights into their herbicidal activity and potential biotechnological production. Through genomic analysis, a cryptic biosynthetic gene cluster was identified, leading to a better understanding of the mechanism behind targeting the plant TORK pathway. The existence of other variants in plant-associated Streptomyces strains raises new questions about the function of this lipopeptide.

Summary: In this study, vanillin was found to efficiently catalyze hydrogen peroxide to produce reactive radicals and accelerate the chemiluminescence signal of luminol-hydrogen peroxide. By combining with beta-CD, the chemiluminescence intensity was further enhanced. Based on the catalytic and stabilizing functions, a low-cost, rapid, and simple chemiluminescence sensor was developed for the detection of vanillin.

Summary: The study demonstrates the heterogeneous gold(I)-catalyzed oxidative ring expansion of alkynyl quinols using a benzyldiphenylphosphine-modified MCM-41-immobilized gold(I) complex as the catalyst and 8-methylquinoline N-oxide as the oxidant, resulting in the formation of functionalized tropone derivatives in good to excellent yields. The methodology can also be extended to construct other seven- or six-membered ring systems, such as dibenzotropones, dibenzooxepines, phenanthrenes, and quinolin-2(1H)-ones, with the new heterogeneous gold(I) complex showing recyclability up to eight times without loss of catalytic efficiency.

Summary: The application of amino acids and small peptides as asymmetric organocatalysts in aldol and conjugate addition reactions has been a significant breakthrough in the past decade. Desired stereoselectivity in these reactions can be achieved through the tunable diversity elements present in amino acids/peptides. The literature is classified based on the use of single amino acids or peptides as catalysts, with detailed information on the design and recent applications of these catalysts.

Summary: An efficient method for the synthesis of C-1 aryl enones from aryl iodides and glycal enones through palladium-catalyzed cross-coupling reactions under ligand-free conditions was developed. This method is applicable to a wide range of aryl iodides with different electron-donating and withdrawing groups, and can be carried out under mild reaction conditions.

Summary: A series of tetrahydrobenzo[b]pyran derivatives were successfully synthesized through three-component reactions, with high yields and relatively shorter reaction times. The optimal reaction conditions were found to be a water-ethanol solvent system, a reaction temperature of 60(o)C, and a 5 mol% loading of the imidazolium catalyst. This method exhibits operational simplicity, no need for chromatographic purification, and avoidance of hazardous solvents.

Summary: The revival of peptide-based drugs has increased the demand for large-scale synthesis, leading to the successful synthesis of a pentapeptide POSS-thioester with high yield and purity using mercapto-functionalized POSS as a soluble support. This method offers a promising way for the scale-up chemical synthesis of peptide thioesters, peptides, peptide amides, cyclic peptides, and even proteins.

Summary: Peptide-based opioid ligands are potential candidates for developing safer and more effective analgesics. In this study, a cyclic pentapeptide library was screened and a novel cyclic peptide, CycloAnt, was identified. CycloAnt exhibited dose- and time-dependent antinociception mediated by MOR and showed lower adverse effects and dependence compared to morphine.
