Summary: Here, we report an unprecedented protocol using ruthenium-catalyzed annulation for the synthesis of 6H-chromeno[4 ',3 ':4,5]imidazo[1,2-a]pyridin-6-one, and a new method for intramolecular chelation-assisted C-H activation to produce functionalized 2-(3-formylimidazo[1,2-a]pyridin-2-yl)phenyl acetate. Furthermore, a one-pot approach using ruthenium catalysis and formic acid has been developed for the gram-scale synthesis of bis(2-phenylimidazo[1,2-a]pyridin-3yl)methane (BIP) and the late-stage functionalization of zolimidine, a marketed drug, with good yield.

Summary: Peptidomimetics with a substituted imidazo[1,2-a ]pyridine fragment were synthesized using a combination of Groebke-Black-burn-Bienayme and Ugi reactions. The resulting products contained substituted imidazo[1,2-a]pyridine and peptidomimetic moieties as pharmacophores, with four diversity points introduced from readily available starting materials. A focused compound library consisting of 20 Ugi products was prepared and screened for antibacterial activity.

Summary: A rapid method for preparing a series of N-heteroarene fluorophores via palladium-catalyzed direct oxidative reactions has been developed, showing color-tunable emissions and high fluorescence quantum yields. The photophysical properties-structure relationship was systematically investigated, leading to the discovery of fluorophores with emissions ranging from 431-507 nm in CH2Cl2 and fluorescence quantum yields up to 91% in CH2Cl2.

Summary: An efficient iodine-catalyzed method for synthesizing imidazo[1,2-a]pyrazines and imidazo[1,2-a]pyridines via one-pot three-component condensations has been reported. The photophysical properties of these new fluorescent derivatives are also presented. The synthesized compounds were evaluated for their anti-cancer activities and one compound showed promising results.

Summary: A general and straightforward method utilizing N,N-dimethylaniline as the methylenation source has been reported for the regioselective construction of C-3 heteroaryl-containing imidazo[1,2-a]pyridines via cross-dehydrogenative coupling under transition-metal-free conditions. The reaction proceeds through a radical pathway and affords C(sp(2))-C(sp(3)) functionalized products in good to excellent yields.

Summary: New imidazo[1,2-a]pyridine candidates were designed by synthesizing derivatives using 2-amino pyridine, TOSMIC, and various aldehydes, showing antioxidant and antifungal potential. The highest antifungal inhibition was observed in compound 12 against Aspergillus fumigatus and Candida albicans, with confocal images and UPLC analysis revealing the antifungal mechanism. In silico studies confirmed the interaction of the synthesized compounds with the active site of the target enzyme, while ADME analysis indicated drug-likeliness of the compounds.

Summary: The annulation of imidazo[1,2-a]pyridines with different substrates under various conditions leads to the formation of different fused products, with the phenyl ring at the C-2 position of the imidazo[1,2-a]pyridines playing a more crucial role in the reaction than alkyl groups. Both transformations follow the ionic mechanism by Pd-catalyzed double C-H bond activation, resulting in highly fluorescent characteristics in the annulated products.

Summary: In this study, a series of novel fluorescent Imidazo [1,2-a] pyridines (IMPs) (4a-4p) were synthesized via an eco-friendly microwave-assisted Groebke-Blackburn-Bienayme reaction (GBBR) with good to excellent yields (80-92%) and a green catalyst. These target molecules exhibited absorption in the UV-vis region and emitted blue-to-greenish fluorescence with large Stokes shifts (6780-9011 cm-1) due to an intramolecular charge transfer (ICT) process. Time-dependent density functional theory (DFT) calculations were performed to analyze the structure-property relationships and confirm the ICT characteristics and energy band gaps. Furthermore, imaging studies on HEK293 and HeLa cell lines demonstrated that IMP 4e, which had the highest fluorescence quantum yield (phi fl) of 66%, served as a specific blue fluorescent probe for staining the DNA of the nucleus.

Summary: A selectfluor-promoted oxidative coupling reaction was developed for the formation of C-O, C-N, C-S, and C-Se bonds by reacting quinoxalin-2(1H)-ones with alcohols, amines, thiols, and selenols. The protocol showed high yields (53-95%) and allowed for the decoration of a wide range of quinoxalin-2(1H)-ones at the C3-position with various functional groups. The reaction proceeds via a radical pathway and features mild reaction conditions, broad substrate scope, and metal-and photocatalyst-free conditions.

Summary: This study reports a novel antileishmanial agent C17, which can poison topoisomerase 1-DNA covalent complexes (LdTop1cc) inside parasites and inhibit its religation activity. C17 is effective against both drug sensitive and antimony-resistant L. donovani isolates without affecting human Top1. It overcomes the chemical instability of camptothecin (CPT) and shows promising results in clearing Leishmania parasites in infected mice.

Summary: This study presents a catalyst-dependent regioselective oxidative annulation of 2-arylimidazo[1,2-a]pyridines with cinnamaldehyde derivatives to construct fused N-heterocyclic frameworks. The annulation reaction using [RhCp*Cl-2](2) as catalyst afforded 5-arylnaphtho[1',2':4,5]imidazo[1,2-a]pyridine-6-carbaldehydes, while Pd(OAc)(2) catalyst led to the formation of 1,7-diarylimidazo[5,1,2-cd]indolizine-6-carbaldehydes. The reaction exhibited high yields, broad substrate scope, and excellent functional group tolerance, providing valuable compounds with different functionalities.
