Brain injury and neuronal loss leads to an inflammatory response, which is initiated by the innate immune system. To what extent this immune response is beneficial or detrimental for neurogenesis and regeneration is unclear. We addressed this question during regeneration of dopamine neurons in the adult salamander brain. In contrast to mammals, ablation of dopamine neurons evokes robust neurogenesis leading to complete histological and functional regeneration within four weeks in salamanders. Here we show that similarly to mammals, ablation of dopamine neurons causes microglia activation and an increase in microglia numbers in the ablated areas. Furthermore, microglia numbers remain elevated compared to the uninjured brain at least six weeks after ablation. Suppression of the microglia response results in enhanced regeneration, concomitant with reduced death of dopamine neurons during the regeneration phase. Thus neuroregeneration is not dependent on the absence of an innate immune response, but the suppression of this response may be a means to promote neurogenesis in the adult vertebrate brain. Summary: This study developed a tissue-clearing protocol, CUBIC-f, optimized for fragile brain samples, and demonstrated its effectiveness in quantifying neuronal cell density and characterizing neural progenitor cells. CUBIC-f was found suitable for preserving tissue integrity in embryonic mouse brains, larval, and adult salamander brains compared to traditional CUBIC and CLARITY protocols.

Locomotion dependent neuron-glia interactions control neurogenesis and regeneration in the adult zebrafish spinal cord

Weipang Chang, Andrea Pedroni, Maria Bertuzzi, Caghan Kizil, Andras Simon, Konstantinos Ampatzis

Summary: The study shows that physical exercise can activate neurogenesis in the spinal cord of adult zebrafish through cholinergic neurotransmission and a reduction of GABA(A) receptors. The researchers also found that functional recovery after spinal cord injury can be promoted by stimulating cholinergic receptors and interfering with GABAergic signaling. The findings suggest a model for activity-dependent neurogenesis in the adult zebrafish spinal cord during homeostasis and regeneration.

Tig1 regulates proximo-distal identity during salamander limb regeneration

Summary: Through single-cell RNAseq analysis, Tig1 has been identified as a central cell surface determinant of proximal identity in salamander limb regeneration. Its overexpression leads to regeneration defects in the distal elements and elicits proximal displacement of blastema cells, while its neutralisation blocks proximo-distal cell surface interactions.

Modelling Alzheimer's disease using human brain organoids: current progress and challenges

Mario Yanakiev, Olivia Soper, Daniel A. Berg, Eunchai Kang

Summary: Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized by gradual memory loss and declining cognitive and executive functions. The cause of AD is not well understood and there is no effective cure. Human brain organoids provide unique opportunities to study AD and its pathological components, filling the gap left by traditional model systems.

Neural correlates of social interaction anxiety and their relation to emotional intelligence: A resting-state fMRI study

Song Xue, Feng Kong, Yiying Song, Jia Liu

Summary: This study used resting-state functional magnetic resonance imaging to explore the relationship between individual's spontaneous neural activity and social interaction anxiety in a nonclinical population. The results showed that social interaction anxiety was correlated with the fractional amplitude of low-frequency fluctuations in several brain regions, and that emotional intelligence partially mediated this relationship. This study provides evidence for the neural basis of social interaction anxiety in the normal population and highlights the role of emotional intelligence in this anxiety.

Development of the human medullary arcuate nucleus from mid-gestation to the perinatal period: A morphometric study

Katsuyuki Yamaguchi, Takuya Yazawa

Summary: This study provides morphometric data on the development of the human medullary arcuate nucleus (AN) by examining the brains of preterm and perinatal infants. The results show that AN morphology demonstrates asymmetry and individual variability during the fetal period. The volume and neuronal number of AN increase exponentially with age, while neuronal density decreases exponentially. The AN may undergo neuron death and neuroblasts production after mid-gestation.

Transfer of massive mitochondria from astrocytes reduce propofol neurotoxicity

Zhan Zhou, Weixin Dai, Tianxiao Liu, Min Shi, Yi Wei, Lifei Chen, Yubo Xie

Summary: Studies have shown that propofol-induced neurotoxicity is caused by disruption of mitochondrial fission and fusion, leading to an energy supply imbalance for developing neurons. Healthy mitochondria released by astrocytes can migrate to compromised neurons to mitigate propofol-induced neurotoxicity, but the exact mechanisms involved still need further clarification.

Alamandine injection in the periaqueductal gray and rostral ventromedial medulla attenuates allodynia induced by sciatic nerve ligation in rats

Summary: This study investigated the effects of alamandine on allodynia in a rat model and found the presence of MrgD receptors in the vlPAG and RVM regions. Microinjection of alamandine resulted in a significant increase in paw withdrawal threshold and could be blocked by an MrgD receptor antagonist. Upregulation of MrgD receptor expression following allodynia induction suggests a potential compensatory mechanism in response to pain.

Evaluation of the separate and combined effects of anodal tDCS over the M1 and F3 regions on pain relief in patients with type-2 diabetes suffering from neuropathic pain

Ahmad Alipour, Roghayeh Mohammadi

Summary: The present study aimed to investigate the separate and combined effects of anodal transcranial direct current stimulation (tDCS) over the primary motor cortex (M1) and left dorsolateral prefrontal cortex (F3) regions on pain relief in patients with type-2 diabetes suffering from neuropathic pain (NP). The results showed that tDCS had the potential to induce pain relief in patients with type-2 diabetes suffering from NP. The mean perceived pain intensity in the posttest was lower in the M1 stimulation group than in the F3 stimulation group. However, more trials with larger sample sizes are necessary to define clinically relevant effects.

The cognitive architecture of verbal humor in schizophrenia

Summary: This study found a general impairment in humor comprehension in individuals with schizophrenia, with mental jokes being more difficult for both patients and controls. Humor comprehension was closely associated with the patients' overall pragmatic and linguistic profile, while the association with Theory of Mind (ToM) was minimal. Another notable finding was the increased appreciation of humor in individuals with schizophrenia, who rated jokes as funnier than controls did, regardless of whether they were correctly or incorrectly completed. The funniness ratings were not predicted by any measure, suggesting a dimension of humor untied to cognition or psychopathology.
