TPH1 218A/C polymorphism is associated with major depressive disorder and its treatment response

The association between the tryptophan hydroxylase 1 (TPH1) 218A/C polymorphism and (1) severity of major depressive disorder (MDD) and (2) response to treatment was studied. There were three study populations, the first consisting of 119 treatment-resistant MDD inpatients treated with electro-convulsive therapy (ECT), and the second of 98 MDD open care patients treated with selective serotonin reuptake inhibitors (SSRI). In addition, there was a control population of 395 healthy blood donors. The first aim of the study was to compare the genotypes of the patient with those of the healthy controls and between patient populations. The second aim was to compare the genotypes of MDD patients achieving remission with basic SSRI treatment (MADRS < 8) with the genotypes of non-responders to ECT (defined as MADRS > 15). TPH1 218A/C polymorphism was associated with the risk of MDD. CC genotype was significantly more common in patients (including both ECT and SSRI treated patients) than in controls (38.2% and 26.8% respectively; p = 0.008), and its frequency was significantly higher in more severe forms of depression, i.e. in ECT treated patients compared with SSRI treated patients (42.0% and 33.7%, p = 0.026). CC genotype was also associated with lower probability of achieving remission. It was significantly more frequent among ECT non-responders than among SSRI remitters (53.1% and 23.3%, p = 0.049). In this Finnish population TPH1 218A/C polymorphism was associated with the risk of MDD and treatment response; CC genotype was associated with the increased risk of MDD and lower probability of responding treatment. Parenthood and changes in physical activity from early adulthood to mid-life among Finnish adults

Summary: This study examined the long-term associations between parenthood-related factors and physical activity from young adulthood to midlife. Results showed that parents with children under 6 years old were less likely to be physically active than those without children. However, there was no difference in meeting the aerobic physical activity recommendation between parents with older children and childless individuals. The older the youngest child, the more likely the parents were to be physically active.

Summary: Refractive error is a complex eye condition caused by genetic and environmental factors. Genome-wide association studies have identified common genetic risk factors, but a significant portion of the heritability is still unexplained. By conducting gene-based association tests, we identified 129 unique genes associated with refractive error in a large dataset. Some of the novel candidates, such as PDCD6IP, PER3, and P4HTM, show promise for future functional studies and validation.

Stress-Related Exhaustion, Polygenic Cognitive Potential, and Cognitive Test Performance - A General Population Study

Summary: This study aimed to investigate the relationship between stress-related exhaustion (chronic or short-term, and co-occurring with depression or not) and cognitive performance, and whether polygenic cognitive potential modifies these associations. The study found that high stress-related exhaustion, especially chronic, was associated with slower reaction time. Clinical levels of depression were only related to slower reaction time. Polygenic cognitive potential did not modify these associations. There were no differences in cognitive performance between individuals with co-occurring exhaustion and depression vs. those with only either condition.

Summary: Through meta-analysis of epigenome-wide association studies, we found significant associations between dietary and supplemental intake of vitamins C and E with DNA methylation. Vitamin C intake was associated with 4,656 CpG sites, while vitamin E intake was associated with 160 CpG sites. Vitamin C intake was found to be related to systems development and the immune response, while no significant associations were found for vitamin E intake with any biological pathways investigated.

Summary: This observational study examines the complex temporal associations between body-mass index (BMI), waist-hip ratio (WHR), and circulating metabolomics. It uses longitudinal and cross-sectional population-based datasets and new systems epidemiology tools. The results show that age-associated weight variation in adults between 24 and 49 manifests as temporal divergence in BMI and uniform progression of WHR.

Summary: This study used data from 2,325 population-based studies to analyze the height and BMI of children and adolescents aged 5-19 years in urban and rural areas from 1990 to 2020. The results showed that the height advantage of urban residents over rural residents has decreased in most countries, while it has amplified in sub-Saharan Africa. The BMI difference between urban and rural areas is small in most countries, with faster BMI growth in urban areas observed in some regions.

Summary: Through a large-scale multi-ancestry genome-wide association meta-analysis, 1020 independent association signals implicating 559 genes related to COPD were identified. These genes were enriched in 29 pathways. Variants showed heterogeneity across ancestries, age and smoking groups, and the genetic risk score was strongly associated with COPD across ancestry groups. New potential causal variants, genes, proteins and pathways were highlighted, including those targeted by existing drugs. These findings contribute to a better understanding of lung function and COPD mechanisms and have implications for functional genomics experiments and future COPD therapies.

Polygenic liabilities underlying job stress and exhaustion over a 10-year follow-up: A general population study

Summary: We investigated whether individuals with a high genetic loading for schizophrenia and major depression (PGL) but without the respective disorders were still susceptible to milder forms of ill-being in terms of job strain or exhaustion. Using data from the Young Finns Study, we found that high PGL did not predict less favorable perceptions of job environment, but it did predict a higher trajectory of exhaustion in early adulthood and middle age. Additionally, high PGL predicted a stronger increase in exhaustion at higher levels of job strain, even after controlling for various factors.

Neural correlates of social interaction anxiety and their relation to emotional intelligence: A resting-state fMRI study

Summary: This study used resting-state functional magnetic resonance imaging to explore the relationship between individual's spontaneous neural activity and social interaction anxiety in a nonclinical population. The results showed that social interaction anxiety was correlated with the fractional amplitude of low-frequency fluctuations in several brain regions, and that emotional intelligence partially mediated this relationship. This study provides evidence for the neural basis of social interaction anxiety in the normal population and highlights the role of emotional intelligence in this anxiety.

Development of the human medullary arcuate nucleus from mid-gestation to the perinatal period: A morphometric study

Summary: This study provides morphometric data on the development of the human medullary arcuate nucleus (AN) by examining the brains of preterm and perinatal infants. The results show that AN morphology demonstrates asymmetry and individual variability during the fetal period. The volume and neuronal number of AN increase exponentially with age, while neuronal density decreases exponentially. The AN may undergo neuron death and neuroblasts production after mid-gestation.

Summary: The present study aimed to investigate the separate and combined effects of anodal transcranial direct current stimulation (tDCS) over the primary motor cortex (M1) and left dorsolateral prefrontal cortex (F3) regions on pain relief in patients with type-2 diabetes suffering from neuropathic pain (NP). The results showed that tDCS had the potential to induce pain relief in patients with type-2 diabetes suffering from NP. The mean perceived pain intensity in the posttest was lower in the M1 stimulation group than in the F3 stimulation group. However, more trials with larger sample sizes are necessary to define clinically relevant effects.

The cognitive architecture of verbal humor in schizophrenia

Summary: This study found a general impairment in humor comprehension in individuals with schizophrenia, with mental jokes being more difficult for both patients and controls. Humor comprehension was closely associated with the patients' overall pragmatic and linguistic profile, while the association with Theory of Mind (ToM) was minimal. Another notable finding was the increased appreciation of humor in individuals with schizophrenia, who rated jokes as funnier than controls did, regardless of whether they were correctly or incorrectly completed. The funniness ratings were not predicted by any measure, suggesting a dimension of humor untied to cognition or psychopathology.
