Summary: We investigated the effects of histamine and GABA release from TMN axons projecting to the PFC on circuit processing. Histamine stimulated fast-spiking interneurons, while released GABA enhanced tonic inhibition on PyrNs. Histamine-induced gain changes were blocked by histamine receptor antagonists, while GABA-induced gain changes were blocked by GABAA receptor antagonists. The GABA modulation of PyrN excitability by TMNHDC axons increased significantly in older mice, potentially enhancing information processing and maintaining cognition in aging individuals.

Summary: Itch is a complex sensation regulated by neural circuits involving the nucleus accumbens (NAc) and excitatory afferents from the medial prefrontal cortex (mPFC), amygdala, and hippocampus. This study reveals that acute itch decreases the intrinsic membrane excitability in dopamine receptor D1-expressing medium spiny neurons (Drd1-MSNs) in the NAc core, and that chemogenetic activation of Drd1-MSNs alleviates itch-induced scratching behaviors. Furthermore, it was found that excitatory synaptic afferents from the mPFC play a crucial role in itch modulation.

Summary: Cues in the environment can affect fear and reward-seeking behaviors, with safety cues reducing fear. Previous research showed that the infralimbic prefrontal cortex plays a role in suppressing fear during safety cues. This study aimed to investigate safety cue-specific neural activity in the infralimbic cortex using a Pavlovian conditioning paradigm. The results showed that neurons were excited by both fear and safety cues, with some neurons being excited by both fear and reward cues and some showing bidirectional responses. Neural activity was also negatively correlated with freezing behavior. These findings emphasize the importance of the infralimbic cortex in encoding specific aspects of conditioned inhibitors during active fear suppression.

The findings suggest that Drd2 regulates synaptic pruning through cell-autonomous mechanisms involving mTOR signaling, rather than synapse formation. Deficits in Drd2-mediated synaptic pruning during adolescence may lead to hyper-glutamatergic function and anxiety-like behaviors in adulthood.

Summary: Chemogenetic activation of a specific subset of pyramidal cells in the mPFC to DS pathway drives running behavior specifically during periods of limited food availability, leading to a causal role of this pathway in the decision to run during food scarcity. Suppression of these cells reduces running response to food restriction, with distinct characteristics observed in mPFC-to-DS cells compared to neighboring non-DS-projecting cells. These findings suggest a potential role for enhancing PFC-to-DS activity in influencing neural circuits under food restriction conditions.

The NMDA antagonist drug memantine shows potential in attenuating the neurotoxicity caused by cocaine and HIV.

Summary: Alcohol use disorder is associated with changes in the medial prefrontal cortex, including altered glutamatergic transmission and deficits in executive functions. Acamprosate can reduce alcohol craving and relapse by improving alcohol-induced dysregulation of glutamatergic signaling. This study found that acamprosate and its active moiety calcium can improve deficits in cognitive flexibility and altered glutamatergic signaling in mice that self-administered alcohol under goal-directed conditions. However, under conditions that bias behavior towards habitual responding, the effects of acamprosate and calcium on glutamatergic transmission were not observed.
