Relations between brain tissue loss, CSF biomarkers, and the ApoE genetic profile: a longitudinal MRI study

Previously it was reported that Alzheimer's disease (AD) patients have reduced beta amyloid (A beta(1-42)) 1 and elevated total tau (t-tau) and phosphorylated tau (p-tau(181p)) in the cerebrospinal fluid (CSF), suggesting that these same measures could be used to detect early AD pathology in healthy elderly individuals and those with mild cognitive impairment (MCI). In this study, we tested the hypothesis that there would be an association among rates of regional brain atrophy, the CSF biomarkers A beta(1-42), t-tau, and p-tau(181p) and apolipoprotein E (ApoE) epsilon 4 status, and that the pattern of this association would be diagnosis-specific. Our findings primarily showed that lower CSF A beta(1-42) and higher tau concentrations were associated with increased rates of regional brain tissue loss and the patterns varied across the clinical groups. Taken together, these findings demonstrate that CSF biomarker concentrations are associated with the characteristic patterns of structural brain changes in healthy elderly and mild cognitive impairment subjects that resemble to a large extent the pathology seen in AD. Therefore, the finding of faster progression of brain atrophy in the presence of lower A beta(1-42) levels and higher tau levels supports the hypothesis that CSF A beta(1-42) and tau are measures of early AD pathology. Moreover, the relationship among CSF biomarkers, ApoE epsilon 4 status, and brain atrophy rates are regionally varying, supporting the view that the genetic predisposition of the brain to beta amyloid and tau mediated pathology is regional and disease stage specific. Summary: The study evaluated the role of cortical amyloid deposition in memory dysfunction and dementia risk associated with late-life depression. Contrary to expectations, the late-life depression group showed less A13 deposition than the non-depressed group, and A13 deposition was not associated with depression history characteristics. However, A13 deposition was associated with memory performance.

Summary: In this study, a multilabel non-ADNC classifier using MRI signatures was developed for the identification of non-Alzheimer's disease neuropathological changes. The model showed high accuracy in distinguishing TDP-43, LBD, and CAA, outperforming reference models without MRI and ADNC biomarkers. Considering non-ADNC decreased the required sample size to detect cognitive decline.

Network-constrained technique to characterize pathology progression rate in Alzheimer's disease

Summary: Current methods for measuring the chronic rates of cognitive decline and degeneration in Alzheimer's disease rely on longitudinal neuropsychological batteries and clinical neuroimaging. Predicting future disease progression has proven challenging, with brain network diffusion models offering a potential solution to accurately estimate pathology transmission rates. Using machine learning, a new metric based on MRI-derived atrophy data was able to predict disease progression across different diagnostic groups and genetic risk factors.

Auditory robustness and resilience in the aging auditory system of the desert locust

Summary: This study investigated the effects of age on the robustness and resilience of auditory system using the desert locust. The researchers found that gene expression changes were mainly influenced by age rather than noise exposure. Both young and aged locusts were able to recover their auditory nerve function within 48 hours of noise exposure, but the recovery of transduction current magnitude was impaired in aged locusts. Key genes responsible for robustness to noise exposure in young locusts and potential candidates for compensatory mechanisms in auditory neurons of aged locusts were identified.
