Evidence for Two Modes of Binding of the Negative Allosteric Modulator SB269,652 to the Dopamine D2 Receptor

Summary: SB269,652 is the first negative allosteric modulator of the dopamine D-2 receptor, but its mechanism of action is not fully understood. This study investigated the interaction between dopamine and SB269,652 and found that the inhibitory potency of SB269,652 is greatly increased after ligand pre-incubation. Mutagenesis experiments suggested that specific amino acids in the orthosteric binding site and the secondary binding pocket are involved in this effect. These findings expand our knowledge of how SB269,652 competes with dopamine at the D2R and could be important for the development of new D2R ligands.

Summary: This study found that intranasal infusion of GALR2 and Y1R agonists promotes neuroblast proliferation and induces the expression of the neurotrophic factor BDNF in the ventral hippocampus, resulting in antidepressant effects. These effects may be mediated by increased formation of Y1R-GALR2 heteroreceptor complexes. These findings may provide a new approach for the development of novel therapies for major depressive disorder.

Dopamine-induced arrestin recruitment and desensitization of the dopamine D4 receptor is regulated by G protein-coupled receptor kinase-2

Summary: In this study, a novel nanoluciferase complementation assay was used to investigate the dopamine-induced recruitment of beta-arrestin2 (beta arr2) and G protein-coupled receptor kinase-2 (GRK2) to the D4 receptor. The desensitization of D4 receptor-evoked G protein-coupled inward rectifier potassium (GIRK) current responses was also studied. The results showed that coexpression of GRK2 enhanced the potency of dopamine to induce beta arr2 recruitment to the D4 receptor and accelerated the rate of desensitization of D4 receptor-evoked GIRK responses.

Unique effect of clozapine on adenosine A2A-dopamine D2 receptor heteromerization

Summary: This study investigates the effect of three typical antipsychotic drugs on A2AR-D2R heteromer formation in mammalian cells using a NanoBiT assay. It was found that haloperidol and aripiprazole significantly increase the density of A2AR-D2R heteromers after 16 hours of exposure, while clozapine does not. Conversely, clozapine leads to a significant decrease in A2AR-D2R heteromerization after 2 hours of treatment, whereas haloperidol and aripiprazole do not. Computational binding models reveal distinct molecular signatures that explain these differential effects on heteromerization.

5HT1AR-FGFR1 Heteroreceptor Complexes Differently Modulate GIRK Currents in the Dorsal Hippocampus and the Dorsal Raphe Serotonin Nucleus of Control Rats and of a Genetic Rat Model of Depression

Summary: This study investigates the effects of 5HT1AR-FGFR1 complex activation on hippocampal pyramidal neurons and midbrain dorsal raphe serotonergic neurons using an electrophysiological approach. The results show that in normal rats, FGFR1 agonist-induced allosteric action counteracts the ability of 5HT1AR to open GIRK channels, leading to increased neuronal firing. However, in a depression rat model, the change in 5HT1AR caused by FGFR1 does not affect the GIRK channels, except in CA2 neurons. Therefore, the reduced inhibitory ability of FGFR1 on 5HT1AR-mediated GIRK channel opening may be associated with depression.

Type I interferon shapes brain distribution and tropism of tick-borne flavivirus

Summary: The authors use a multimodal imaging-snRNAseq transcriptomics strategy to study the distribution of a neurotropic tick-borne flavivirus in the brain. They find that absence of interferon signaling increases infection of resident microglia. Using multimodal imaging and single nuclei RNA-sequencing, they visualize viral pathogenesis and define molecular mechanisms. The study highlights the importance of understanding viral tropism and the role of immune response in neurotropic flavivirus infection.

An E280K Missense Variant in KCND3/Kv4.3-Case Report and Functional Characterization

Summary: A five-year-old girl with headache attacks and gait disturbances was found to have a new variant KCND3 c.838G>A, p.E280K in her father and sisters, but not in her mother and brother. KCND3 encodes voltage-gated potassium channel D3 (Kv4.3) and mutations have been associated with spinocerebellar ataxia type 19/22 (SCA19/22) and cardiac arrhythmias. The family members did not exhibit ataxia or parkinsonism, but both sisters showed mild developmental delay. Functional experiments showed that E280K affects channel activation and inactivation. Overall, E280K is associated with developmental delay but appears to be relatively benign.

In Vitro Comparison of Ulotaront (SEP-363856) and Ralmitaront (RO6889450): Two TAAR1 Agonist Candidate Antipsychotics

Summary: The study compared the efficacy of ulotaront and ralmitaront at TAAR1, 5-HT R-1A, and dopamine D 2. It was found that ralmitaront had lower efficacy at TAAR1 and had no activity at 5-HT R-1A and dopamine D 2. Compared to ulotaront, ralmitaront had lower efficacy and slower kinetics at TAAR1 and lacked efficacy at 5-HT R-1A.
