Summary: High expression levels of alpha-SMA and hypoxia markers are associated with poor prognosis in pancreatic cancer patients. Mechanistically, hypoxia induces HGF expression and secretion in PSC via HIF-1 alpha, which then activates Met and suppresses pancreatic cancer cell sensitivity to EGFR inhibitors. The combination of EGFR inhibitor and Met inhibitor shows promise for pancreatic cancer treatment based on the demonstrated signaling axis between PSC and cancer cells.

Summary: Some flavonoids have been found to enhance the production of EPO (erythropoietin) by increasing the activity of hypoxia response element (HRE). This suggests that flavonoids may have potential as important components of food supplements for brain health.

Summary: Cardiovascular diseases are the leading cause of death worldwide, and hypoxia-inducible factors (HIFs) play an important role in their pathogenesis. Recent studies have revealed the role of cell-specific HIFs in various cardiovascular diseases, but the potential clinical application of HIF inhibitors in the treatment of cardiovascular diseases is not well understood.

Summary: Using genetic data, a Mendelian randomization analysis was conducted to explore the causal relationships between inflammatory cytokines and migraine. The results showed a positive association between hepatocyte growth factor (HGF) and migraine risk, while interleukin-2 (IL-2) was considered a downstream consequence of migraine.

Summary: Bone marrow hematopoiesis is regulated by various factors, including bone components, EPO, and FGF23. The interaction between these factors involves HIF pathway, direct stimulation of FGF23 synthesis, and cleavage of FGF23. Recent evidence suggests a complex relationship between FGF23, klotho, EPO, and HIF in hematopoiesis.

Summary: This study found that C3H10T1/2 cells possess the property of HIF2 alpha-dependent Epo expression, and TGF-beta 1 induces both myofibroblast transition and a repressive effect on the Epas1-Epo axis in these cells. The repressive effect of TGF-beta 1 on the Epas1-Epo axis was also confirmed in REP pericytes in vivo, suggesting that inhibition of TGF-beta 1-ALK5 signaling could be a novel treatment approach to rescue EPO expression in REP pericytes of injured kidneys.

Summary: This study demonstrates that hypoxia upregulates the expression of GPT2 in GBM cells through HIF-2 activation. GPT2 is localized in the nucleus and mitochondria and can decrease α-ketoglutarate levels in GBM cells. Inhibition of GPT2 can reduce GBM cell growth and migration, and knockout of GPT2 inhibits GBM tumor growth in mice.

Summary: Hypoxia is a common pathological condition induced by various events, with the body's adaptation to it being crucial for health and disease. HIFs, a family of transcription factors, play a key role in cellular responses to hypoxia and are increasingly viewed as potential targets for treating a range of hypoxia-associated diseases. The role of HIFs in adaptation to hypoxia is universal across tissue types, including the CNS, where they are involved in regulating neurogenesis, nerve cell differentiation, and neuronal apoptosis, potentially offering new therapeutic opportunities.

Summary: PHD inhibitors are effective in treating CKD-associated anemia by stabilizing HIF and increasing erythropoiesis. However, their impact on kidney fibrosis is inconsistent due to the context-dependent effects of HIF. Studies on pericytes suggest that HIF stabilization in these cells may not negatively affect kidney fibrosis despite increased erythropoietin production.

Summary: Stroke is an acute cerebrovascular disease caused by sudden rupture or blockage of blood vessels in the brain, and HIF-1 plays an important role in regulating various pathways in the pathological process. The roles of HIF-1 in stroke are controversial, involving factors such as ischemic time and degree, and its regulatory mechanisms include inflammation, autophagy, oxidative stress, and apoptosis.

Summary: Artificial materials are important for medical devices such as artificial organs and matrices for regenerative medicine. In this study, mitogenic and differentiation-inducible materials are devised via the simple coating of polypeptides, which enable increased cell growth and differentiation through long-lasting signal transduction triggered by bound growth factors. This superior growth and differentiation are attributed to not causing receptor internalization and subsequent downregulation.
