Fragment- and structure-based drug discovery for developing therapeutic agents targeting the DNA Damage Response

Summary: Cancer affects over one-third of the population directly. The DNA Damage Response (DDR) plays a central role in cancer etiology and therapy, with therapeutic approaches targeting DDR advancing through combinatorial treatments and synthetic lethality. While many DDR proteins are undruggable, Fragment- and Structure-Based Drug Discovery (FBDD, SBDD) have led to the development of therapeutic agents and FDA-approved medicines.

Oxidative stress and impaired oligodendrocyte precursor cell differentiation in neurological disorders

Jan Spaas, Lieve van Veggel, Melissa Schepers, Assia Tiane, Jack van Horssen, David M. Wilson, Pablo R. Moya, Elisabeth Piccart, Niels Hellings, Bert O. Eijnde, Wim Derave, Rudy Schreiber, Tim Vanmierlo

Summary: Oligodendrocyte precursor cells (OPCs) play a crucial role in various neurodegenerative disorders, with oxidative/carbonyl stress potentially impacting their differentiation and function, contributing to disease progression. Investigating how oxidative/carbonyl stress disrupts OPC differentiation may offer new directions for therapeutic strategies in neurodegenerative disorders.

FEN1 Blockade for Platinum Chemo-Sensitization and Synthetic Lethality in Epithelial Ovarian Cancers

Katia A. Mesquita, Reem Ali, Rachel Doherty, Michael S. Toss, Islam Miligy, Adel Alblihy, Dorjbal Dorjsuren, Anton Simeonov, Ajit Jadhav, David M. Wilson, Ian Hickson, Natalie J. Tatum, Emad A. Rakha, Srinivasan Madhusudan

Summary: Increased expression of FEN1 in ovarian cancer is associated with poor survival outcomes, while inhibition of FEN1 can enhance platinum sensitivity and exhibit synthetic lethality in BRCA2 and POL beta deficient cells. These findings suggest that FEN1 is a promising target for ovarian cancer therapy.

Oxidative DNA Damage in the Pathophysiology of Spinal Cord Injury: Seems Obvious, but Where Is the Evidence?

Elle E. M. Scheijen, Sven Hendrix, David M. Wilson

Summary: Oxidative stress plays a crucial role in spinal cord injury (SCI), causing damage to proteins, nucleic acids, lipids, cytoskeleton, and organelles. Oxidative DNA damage is likely a major contributor to the pathogenesis of SCI, although the evidence supporting this hypothesis is limited. Improved analysis methods are needed to uncover the role of DNA damage and repair in SCI.
