A series of novel hyperbranched polyselenides and polytellurides with multiple catalytic sites at the branching units has been synthesized via the polycondensation of A2 + B3 monomers. The GPx-like activities of these polymer mimics were assessed and it was found that the polytellurides showed higher GPx-like activities than the corresponding polyselenides. Interestingly, the polymers with higher molecular weights and degree of branching (DB) showed higher GPx-like activities than the analogous lower molecular weight polymer. The enhancement in the catalytical activity of the hyperbranched polymers with increasing molecular weight affirmed the importance of the incorporation of multiple catalytic groups in the macromolecule which increases the local concentration of catalytic sites.

Foldamer-Based Potassium Channels with High Ion Selectivity and Transport Activity

Shuaiwei Qi, Chenyang Zhang, Hao Yu, Jing Zhang, Tengfei Yan, Ze Lin, Bing Yang, Zeyuan Dong

Summary: Small molecules with natural channel-like functions have potential in treating human diseases. Aromatic helical scaffolds were used to develop foldamer-based ion channels of varying sizes, demonstrating the importance of channel size in ion transport selectivity and leading to the discovery of promising artificial K+ and sodiumpreferential channels.

Betulonic Acid Derivatives Interfering with Human Coronavirus 229E Replication via the nsp15 Endoribonuclease

Annelies Stevaert, Besir Krasniqi, Benjamin Van Loy, Tien Nguyen, Joice Thomas, Julie Vandeput, Dirk Jochmans, Volker Thiel, Ronald Dijkman, Wim Dehaen, Arnout Voet, Lieve Naesens

Summary: This study identified a class of HCoV inhibitors targeting nsp15, with lead molecule 5h showing strong inhibitory effects on HCoV-229E replication. However, a mutant virus with nsp15 endoribonuclease active site mutation showed reduced sensitivity to 5h. The findings revealed a druggable pocket in the nsp15 hexamer with relevance for anti-coronavirus drug development.

Summary: This study investigates the relationship between channel size and ion selectivity by developing a new type of artificial ion channel based on pore-forming helical polymers. The artificial channel exhibits high transport activity and Na+ selectivity, and it transports Na+ ions in a different manner compared to natural Na+ channels. Overall, this research provides crucial insights into ion selectivity in the field of ion channels.

Dimensionally confined nanosheets self-assembled through self-shielding multiple hydrogen bonding interactions in aqueous media

Jing Zhang, Shuaiwei Qi, Hao Yu, Ze Lin, Bao Li, Ming Wang, Zeyuan Dong

Summary: In this study, a simple supramolecular strategy was adopted to effectively control the tautomerism of ureidopyrimidinone (UPy) moiety and achieve the complete arrangement of enol configuration. The UPy derivatives containing self-complementary quadruple hydrogen bonding interactions could spontaneously self-assemble into well-controlled, self-organized supramolecular nanostructures in both chloroform and water. This study demonstrates the utility and feasibility of multiple hydrogen bonds for directing the self-assembly of small-molecule building blocks in aqueous media, providing a strategy for constructing well-defined and stable supramolecular architectures with chemical functionalities and physical properties as advanced materials for biological applications.

Lei Zhang, Chenyang Zhang, Xin Dong, Zeyuan Dong

Summary: In this study, artificial supramolecular lithium channels composed of pore-containing small aromatic molecules were developed for the treatment of psychiatric disorders. These channels showed unprecedented transmembrane transport of exogenous lithium ions with high selectivity. The discovery of the basic requirements for ion transport function in these channels may have important implications in studying the physiological role of intracellular lithium ions and treating psychiatric disorders.

Supramolecular 2D monolayered nanosheets constructed by using synergy of non-covalent interactions

Jing Zhang, Shuaiwei Qi, Chenyang Zhang, Wenzhe Wang, Qinwen Ding, Hongyu Zhang, Zeyuan Dong

Summary: A straightforward and rational approach was used to construct supramolecular assemblies with ordered nanostructures in a two-dimensional arrangement. By taking advantage of the synergistic effect of multiple non-covalent interactions, the designed molecular monomer had a specific orientation in the self-assembly process, thus achieving two-dimensional control. The strategy of collaborative self-assembly using multiple non-covalent interactions is expected to be extended to the construction of various unique supramolecular 2D materials.
