In this study, polymer-ceramic composite 3D-printed scaffolds with silk fibroin infill were investigated for their cell adhesion. The presence of silk fibroin improved cell adhesion and viability, but the mechanical strength remained similar.

Summary: A systematic study was conducted to investigate the effects of surface topography on cell adhesion and function, revealing that the geometry and arrangement of submicron pillars have a significant impact on cell adhesion, formation of focal adhesions, and cell morphology. Cells interacting with tall and dense pillars exhibited enhanced adhesion parameters and formation of focal adhesions, as well as specific cell settlement patterns and cytoskeletal organization. Additionally, tall and dense submicron pillars upregulated the expression of osteopontin and promoted matrix mineralization.

Summary: Cancer is a highly heterogeneous disease with (epi)genetic characteristics that determine resistance to therapy. Preclinical research has identified the adhesome as a critical resistance mechanism in multiple cancers. This study connects preclinical data with transcriptomic and patient survival data, revealing a set of overexpressed genes in different cancers that negatively affect patient survival, particularly in radiotherapy-treated cohorts. These genes, including key integrins and their interconnectors, may serve as therapeutic targets in cancer treatment.

Summary: This study compared cfRNA profiles between COVID-19 patients and healthy donors, and found 380 up-regulated cfRNA molecules in all COVID-19 patients. Several potential biomarkers were identified. Antiviral and neutrophil activation-related genes showed decreased expression levels during treatment, which is consistent with enhanced inflammatory response in COVID-19 patients. Differential expression of noncoding RNAs and changes in tRNA pools related to SARS-CoV-2 replication were also observed. Additionally, pneumonia-related microorganisms were detected in the plasma of COVID-19 patients.

Summary: EVs play a critical role in the transport of extracellular small RNA, but little is known about the carriers of cf-mRNA in human biofluids and their association with cancer. In this study, the researchers analyzed the transcriptome of plasma from lung cancer, liver cancer, multiple myeloma, and healthy donors. They found that the majority of cf-mRNA was enriched in EVs, which showed remarkable stability in RNase-rich environments. The specific enrichment patterns of cancer-associated cf-mRNA in each plasma fraction were observed, suggesting the potential of EV subpopulations as a liquid biopsy approach.

Summary: Sequencing studies have revealed differences in gene expression between smouldering multiple myeloma (SMM) and symptomatic multiple myeloma (MM), with shared dysregulated genes highlighting the importance of the PRC2.1 complex. Subgroup-specific differences exist even at the SMM stage of disease, suggesting distinct biological features drive progression within each TC molecular subgroup. The data suggest that MMSET SMM has already transformed, while other precursor diseases are distinct clinical entities from their symptomatic counterpart.

Summary: A Mesenchymal stem cell Differentiation Prediction (MeD-P) framework is reported for predicting biomaterial-induced cell lineage fate. MeD-P integrates public RNA-seq data related to tri-lineage differentiation of human mesenchymal stem cells (hMSCs) and uses a predictive model based on the k-nearest neighbors (kNN) strategy. MeD-P achieves an overall accuracy of 90.63% on testing datasets, significantly higher than the model based on canonical marker genes (80.21%). Furthermore, MeD-P accurately predicts lineage fate on different types of biomaterials as early as the first week of hMSCs culture. In conclusion, MeD-P is an efficient and accurate strategy for stem cell lineage fate prediction and preliminary biomaterial functional evaluation.
