A novel multi-epitope peptide vaccine against cancer: An in silico approach

Cancer immunotherapy has an outstanding position in cancer prevention and treatment. In this kind of therapy, the immune system is activated to eliminate cancerous cells. Multi-epitope peptide cancer vaccines are manifesting as the next generation of cancer immunotherapy. In the present study, we have implemented various strategies to design an efficient multi-epitope vaccine. CD8+ cytolytic T lymphocytes (CTLs) epitopes, which have a pivotal role in cellular immune responses, helper epitopes and adjuvant, are three crucial components of peptide vaccine. CTL. epitopes were determined from two high immunogenic protein Wilms tumor-1 (WT1) and human papillomavirus (HPV) E7 by various servers, which apply different algorithms. CTL epitopes were linked together by AAY and HEYGAEALERAG motifs to enhance epitope presentation. Pan HLA DR-binding epitope (PADRE) peptide sequence and helper epitopes, which have defined from Tetanus toxin fragment C (TTFrC) by various servers, were used to induce CD4+ helper T lymphocytes (HTLs) responses. Additionally, helper epitopes were conjugated together via GPGPG motifs that stimulate HTL immunity. Heparin-Binding Hemagglutinin (HBHA), a novel TLR4 agonist was employed as an adjuvant to polarize CD4+ T cells toward T-helper 1 to induce strong CTL responses. Moreover, the EAAAK linker was introduced to N and C terminals of HBHA for efficient separation. 3D model of protein was generated and predicted B cell epitopes were determined from the surface of built structure. Our protein contains several linear and conformational B cell epitopes, which suggests the antibody triggering property of this novel vaccine. Hence, our final protein can be used for prophylactic or therapeutic usages, because it can potentially stimulate both cellular and humoral immune responses. (C) 2014 Elsevier Ltd. The results demonstrated that low concentrations of nanoparticles can promote cell growth and biofilm formation, while higher concentrations have adverse effects on microalgal communities. The adaptability of cells to nanoparticles is concentration- and exposure time-dependent.

Telmisartan anti-cancer activities mechanism through targeting N-cadherin by mimicking ADH-1 function

Summary: This study aimed to investigate the effects of Telmisartan, a novel N-cadherin antagonist, on cell migration of cancer cells. The results showed that Telmisartan and ADH-1 significantly reduced cell attachment to N-cadherin coated plate in different cancer cell lines. Telmisartan and Docetaxel also showed significant inhibitory effects on cell migration. Moreover, Telmisartan exerted its anti-proliferation and anti-migration effects by targeting antagonistically N-cadherin, suggesting its potential use as an alternative to ADH-1 in enhancing the anticancer effects of Docetaxel.

In silico design and evaluation of a novel mRNA vaccine against BK virus: a reverse vaccinology approach

Summary: In this study, a mRNA vaccine against BK virus (BKV) was designed using a reverse vaccinology approach. Potential immunogenic epitopes were predicted and selected based on several evaluation criteria. The designed vaccine was antigenic, non-toxic, non-allergenic, and had a high population coverage worldwide. Molecular dynamic simulation studies showed strong interactions between the vaccine and toll-like receptor 4 (TLR4). In silico immune simulation analyses demonstrated increased immune responses following repeated exposure to the vaccine.

A framework for relating natural movement to length and quality of life in human and non-human animals

A geometric approach to the evolution of altruism

Summary: Fisher's geometric model is a useful tool for predicting key properties of Darwinian adaptation, and here it is applied to predict differences between the evolution of altruistic versus nonsocial phenotypes. The results suggest that the effect size maximizing probability of fixation is smaller in the context of altruism and larger in the context of nonsocial phenotypes, leading to lower overall probability of fixation for altruism and higher overall probability of fixation for nonsocial phenotypes.

A mathematical framework for the emergence of winners and losers in cell competition

Network-based uncertainty quantification for mathematical models in epidemiology

Summary: This study forecasts the evolution of the COVID-19 pandemic in Germany using a network-based inference method and compares it with other approaches.
