Protein profile in HBx transfected cells: A comparative iTRAQ-coupled 2D LC-MS/MS analysis

The x protein of HBV (HBx) has been involved in the development of hepatocellular carcinoma (HCC), with a possible link to individual genotypes. Nevertheless, the underlying mechanism remains obscure. In this study, we aim to identify the HBx-induced protein profile in HepG2 cells by LC-MS/MS proteomics analysis. Our results indicated that proteins were differentially expressed in HepG2 cells transfected by HBx of various genotypes. Proteins associated with cytoskeleton were found to be either up-regulated (MACF1, HMGB1, Annexin A2) or down-regulated (Lamin A/C). These may in turn result in the decrease of focal adhesion and increase of cell migration in response to HBx. Levels of other cellular proteins with reported impact on the function of extracellular matrix (ECM) proteins and cell migration, including Ca2+-binding proteins (S100A11, S100A6, and S100A4) and proteasome protein (PSMA3), were affected by HBx. The differential protein profile identified in this study was also supported by our functional assay which indicated that cell migration was enhanced by HBx. Our preliminary study provided a new platform to establish a comprehensive cellular protein profile by LC-MS/MS proteomics analysis. Further downstream functional assays, including our reported cell migration assay, should provide new insights in the association between HCC and HBx. (C) 2009 Elsevier B.V. All rights reserved.

Summary: In this study, fermented BSG using Bacillus subtilis WX-17 was found to have higher nutritional components such as amino acids, fatty acids, and Vitamin K(2)MK7. Additionally, higher amounts of short-chain fatty acids were produced after fermentation. The fermentation also had different effects on gut microbiota composition, demonstrating the potential of utilizing BSG as functional food ingredients through microbial fermentation.

Enhancement of the Isomerization Activity and Thermostability of Cellobiose 2-Epimerase from Caldicellulosiruptor saccharolyticus by Exchange of a Flexible Loop

Summary: This study aimed to optimize the catalytic performances of CEs by flexible loop exchange, resulting in mutants with improved isomerization activity and thermostability. Molecular dynamics simulations showed that loop exchange enlarged the entrance of the active site for substrate binding and benefited proton transfer. This work demonstrated the importance of the flexible loop surrounding the active site in determining the isomerization activity and thermostability of CEs.

Administration of xylo-oligosaccharides improves depressive-like behaviour in mice caused by chronic unpredictable mild stress by altering microbiota composition

Summary: This study investigated the modulatory effects of XOS on emotions and behaviors in a mouse model of chronic stress-induced depression. The results showed that XOS could improve depressive-like behavior by modulating neurotransmitters and gut microbiota. These findings suggest that XOS has the potential to be used as a prebiotic for mood disorders.

Development of lymph node metastasis-related prognostic markers in breast cancer

Summary: A prognostic model of BC based on LNM-related genes was established in this study, providing guidance for prognosis evaluation and precise treatment of BC. The genes related to lymph node metastasis in BC are largely unknown and need further exploration. Searching for potential lymph node metastasis-related genes of BC will provide meaningful biomarkers for BC treatment. Our model, based on TCGA-BRCA data, effectively predicted patient outcomes and classified BC patients. The identified feature genes might have a predictive function in immunotherapy. The results of this study provide a new reference for the prognosis and treatment of BC patients with lymph node metastasis.

Lentiviral gene therapy with IGF2-tagged GAA normalizes the skeletal muscle proteome in murine Pompe disease

Summary: Pompe disease is a lysosomal storage disorder characterized by glycogen accumulation in skeletal muscle, leading to profound pathology. This study showed that lentiviral gene therapy with LV-IGF2.GAAco can nearly completely correct disease-associated proteomic changes, supporting its future clinical development as a new treatment option for Pompe disease.

Exoskeleton protein repertoires in decapod crustaceans revealed distinct biomineralization evolution with molluscs

Summary: This study fills the knowledge gap about the matrix proteins involved in the formation of mineralized exoskeletons in decapod crustaceans. By comparing with well-studied molluscan biomineralization proteins, it is proposed that decapod crustaceans evolved novel proteins while sharing some proteins with molluscs. This study provides insights into the evolution and adaptation of crustaceans and molluscs in forming their exoskeletons.
