Summary: Cell-based therapies using PDLSC may be a promising alternative to MSC(M) and MSC(AT) for periodontal regeneration, as PDLSC showed enhanced proliferative and osteogenic potential. PDLSC expressed osteogenic markers and had higher calcium content and upregulated osteogenic/periodontal genes compared to MSC(M) and MSC(AT). CD146 was specifically expressed by PDLSC, which also had a higher proliferative capacity. However, alkaline phosphatase activity did not increase in PDLSC.

Summary: In this study, the effects of e-cigarette exposure on human gingival mesenchymal stem cells (GMSCs) and periodontal ligament stem cells (PDLSCs) were examined through RNA-seq analysis. The results showed that e-cigarette exposure primarily affected immunoregulation and inflammatory responses in GMSCs, and the microenvironment, differentiation, and corticosteroid response in PDLSCs. Additionally, a novel mechanism involving KDM6B-mediated H3K27me3 on enhancers for gene transcription regulation was identified.

Summary: Periodontal tissue regeneration can be achieved by transplanting clumps of mesenchymal stem cells (MSCs) and extracellular matrix (ECM) complexes (C-MSCs) without artificial scaffold. In a rat model, C-MSCs transplantation successfully induced periodontal tissue regeneration, with the donor cells directly involved in tissue formation. This study demonstrates a promising scaffold-free MSCs transplantation strategy for periodontal disease.

Summary: In this study, human induced pluripotent stem cells (iPSCs) were successfully induced into mesenchymal stem cells (MSCs) under xeno-free conditions. The xeno-free-induced MSCs (XF-iMSCs) were found to have regenerative potential in bone and skeletal muscle regeneration. They promoted the regeneration of surrounding host cells through the secretion of soluble factors, and the secretion of peroxidasin and IGF2 also contributed to myotube differentiation.

Summary: This study aimed to investigate the regulatory mechanism of bone marrow-derived mesenchymal stem cell (BM-MSC) differentiation by humoral factors derived from human periodontal ligament (HPL) cells and human gingival fibroblasts (HGFs). The results showed that histone deacetylase (HDAC) expression and histone H3 acetylation were involved in the regulation of BM-MSC differentiation. In clinical applications, regulation of HDAC expression holds potential for effective periodontal regeneration, particularly bone regeneration.
