Non-linear relationship between 5-HT transporter gene expression and frequency sensitivity of 5-HT signals

P>Much evidence suggests that variation in expression of the 5-hydroxytryptamine (5-HT) transporter (5-HTT) is linked to risk of psychiatric illness, but the neurobiological basis of this association is uncertain. In this study, we investigated the impact of variation in 5-HTT expression on subsecond fluctuations in extracellular 5-HT concentrations ([5-HT](o)). Stimulus-evoked [5-HT](o) was detected using fast-scan cyclic voltammetry at carbon-fibre microelectrodes in the substantia nigra in brain slices from 5-HTT knockout (KO) and 5-HTT over-expressing (OE) mice. Compared with wild-type (WT) controls, evoked [5-HT](o) was greater in KO and less in OE mice. In WT controls, evoked [5-HT](o) was frequency-sensitive; however, in both KO and OE mice, evoked [5-HT](o) showed a striking loss of frequency sensitivity. The latter was observed in WT mice after application of a 5-HTT blocker. These data show that while variation in 5-HTT expression modified the peak magnitude of [5-HT](o) evoked by any given stimulus in a gene dose dependent manner, there was a non-linear relationship between 5-HTT expression and frequency sensitivity. Overall, the findings suggest that variation in 5-HTT expression has a marked effect on frequency sensitivity which is a fundamental property of normal 5-HT transmission.

Summary: Neuroimaging has been extensively used to study brain structure and function in individuals with ADHD and ASD. However, there have been limitations in the current literature, such as small sample sizes and heterogeneous methods. To address these limitations, the ENIGMA-ADHD and ENIGMA-ASD working groups were established to collaborate on large-scale neuroimaging projects. The findings so far have shown overlap and unique differences in cortical structures between ASD and ADHD. Ongoing work is exploring other research questions.

Summary: The study identified shared neurobiological processes associated with differences in cortical thickness among multiple psychiatric disorders, suggesting a common role of prenatal development and postnatal functioning of the cerebral cortex. The analysis of data from 145 cohorts revealed that greater expression of genes specific to pyramidal cells was associated with greater group differences in cortical thickness across all 6 disorders.

Summary: This study revealed the regulatory role of CDH13 in inhibitory synapse function by establishing a human neuronal model. By culturing iGABAs in vitro, we found that CDH13 deficiency increases inhibition, thereby reducing the excitatory/inhibitory balance. Through these findings, we identified a potential mechanism by which CDH13 may contribute to the pathophysiology of brain disorders.

Summary: In this study, peripheral blood mononuclear cells from carriers of SLC2A3 copy number variants were successfully reprogrammed into iPSCs and transgene-free cell clones were generated, which showed high expression of pluripotency genes, the ability to differentiate into cells of all three germ layers, and a normal karyotype. These cell lines will be useful for elucidating the role of metabolic alterations in shared pathophysiological processes across organ boundaries.

Summary: Recent advancements in the genetics of neurodevelopmental disorders have identified the transcription factor FOXP2 as a potential risk gene, with its function suggested to be involved in GABAergic signaling that may affect the behavioral phenotypes commonly observed in NDDs. Further research indicates that Foxp2 may regulate locomotor activity via GABAergic signaling, shedding light on potential targetable mechanisms for understanding and treating NDDs.

Summary: A joint analysis of 11 major psychiatric disorders identified four broad factors underlying the genetic correlations among the disorders, along with multiple loci acting on these factors. These findings suggest the presence of genetic overlap among different psychiatric disorders, but also highlight the heterogeneity within each factor. However, a single dimension of genetic risk does not appear to be useful in explaining the biobehavioral correlates or individual variants of psychiatric disorders.

Summary: The interaction between brain serotonin deficiency and environmental adversity leads to excessive aggression in females. Complete inactivation of the gene encoding tryptophan hydroxylase-2 results in the absence of neuronal serotonin synthesis and aggression in male and female mice. Moderate reduction in brain serotonin levels and exposure to stress also lead to increased aggression in male mice. Furthermore, changes in gene expression and myelination-related processes may underlie the molecular mechanisms of aggressive behavior.
