Summary: This study aimed to explore novel therapeutic targets in pulmonary arterial hypertension (PAH) through bioinformatics and experiments. PARM1, TSHZ2, and CCDC80 were identified as potential intervention targets for PAH. The results showed that PARM1 was overexpressed in PAH animal models and primary pulmonary arterial smooth muscle cells, and promoted PAH through the AKT/FOXO3A/PCNA signaling pathway.

Summary: Using a mouse model of FLT3-ITD AML, researchers found that FLT3-ITD AML LSC were enriched within the ST-HSC population and had increased expression of CXCR4. Deletion of CXCL12 from the microenvironment enhanced targeting of AML cells by Flt3-TKI plus chemotherapy treatment, including enhanced LSC targeting. Inhibition of p38 signaling reduced the maintenance of Flt3-ITD AML LSC and enhanced their sensitivity to treatment.

It was found that CXCL12/CXCR4 signaling is specifically required for artery morphogenesis, and a transitioning population expressing Bmp2 was identified as important during mid-gestation.

Therefore, the observed hypogammaglobulinemia in CD19-deficient individuals is likely due to the absence of memory B cells.

Summary: The enteric nervous system (ENS) and peripheral ganglia provide innervation to the gastrointestinal tract, but the molecular mechanisms controlling extrinsic nerve fiber growth are not well understood. This study investigates the role of CXCR4 and CXCL12 in the development of colorectal innervation. It is found that CXCR4 is expressed in nerve fibers arising from the nerve of Remak (NoR) and the pelvic plexus, while CXCL12 is localized to the hindgut and enteric ganglia. Overexpression of CXCL12 enhances axonal projections to the gut from the NoR, while CXCR4 inhibition disrupts nerve fiber extension.
