Zika virus propagation and release in human fetal astrocytes can be suppressed by neutral sphingomyelinase-2 inhibitor GW4869

Zika virus (ZIKV) is a neurotrophic flavivirus that is capable of infecting humans, leading to brain abnormalities during fetal development. The ZIKV infectivity in neural target cells remains poorly understood. Here, we found that ZIKV specifically infected glial fibrillary acidic protein-and S100B-positive primary human astrocytes derived from fetal brains. In contrast, neuron-specific Class III beta-tubulin (TuJ1)-positive neurons in the astrocyte cultures and SOX2-positive neural progenitor cells derived from the fetal brains were less susceptible to ZIKV infection compared with astrocytes. The infected astrocytes released competent viral particles and manifested programmed cell death with a progressive cytopathic effect. Interestingly, ZIKV infection in human fetal astrocytes induced a significant increase of extracellular vesicles (EVs). Treatment with GW4869, a specific inhibitor of neutral sphingomyelinase-2, decreased EV levels, suppressed ZIKV propagation, and reduced the release of infectious virions in astrocytes. Therefore, ZIKV infects primary human fetal astrocytes and the infection can be suppressed by neutral sphingomyelinase-2 inhibitor GW4869. Further investigation into sphingomyelin metabolism and EVs may provide insights to the therapeutic treatment of ZIKV infection.

Spatial Location Aided Fully-Distributed Dynamic Routing for Large-Scale LEO Satellite Networks

Extensive experiments are conducted to verify the effectiveness and advantages of the proposed approach under large-scale LEO satellite networks.

Towards Predictive Forwarding Strategy in Vehicular Named Data Networking

Summary: This paper proposes a novel predictive forwarding strategy (PRFS) to improve the content delivery efficiency in vehicular ad hoc networks. PRFS uses long short-term memory (LSTM) to amend the neighbor table for more accurate neighboring vehicles' positions and selects the next-hop forwarder based on link reliability and distance along the road. Experimental results show that PRFS can reduce the number of forwarded interest packets and data packets by about 21.29% and 25.75%, respectively, and improve the success ratio of satisfied interest packets by 35.1% compared to existing algorithms.

Joint Beam-Hopping Scheduling and Coverage Control in Multibeam Satellite Systems

Summary: Beam-hopping scheduling technology is crucial for improving the performance of resource-constrained multibeam satellite systems. Existing approaches often waste transmission resources due to fixed beam coverage radius. In this study, we propose DeepBeam, a deep reinforcement learning-based algorithm that optimizes beam-hopping scheduling and coverage control by considering time, space, and beam coverage radius. The results of simulations demonstrate that DeepBeam can enhance MBS throughput and reduce packet loss probability.

Characterization and Differentiation of Grain Proteomes from Wild-Type Puroindoline and Variants in Wheat

Summary: This study compared three variants of hard wheat puroindoline (MY26, GX3, and ZM1) with a soft wheat variety (CM605) to identify differentially abundant proteins using proteomic methods. A total of 6253 proteins were identified and quantified via mass spectrometry, and 208 differentially expressed proteins were found between the hard wheat and soft wheat varieties. Cluster analysis divided the proteins into six clusters, with 67 and 41 proteins being more abundant in CM605 and the hard wheat varieties, respectively. Enrichment analysis detected common GO terms, KEGG pathways, and IPR terms across all three comparisons. Additionally, 12 proteins associated with these terms or pathways were found to be differentially expressed in each comparison, including cysteine proteinase inhibitors, invertases, low-molecular-weight glutenin subunits, and alpha amylase inhibitors, which may play a role in grain hardness regulation. The candidate genes identified in this study may contribute to future analyses of the regulatory mechanism underlying grain hardness.

Genome-wide systematic characterization of the NRT2 gene family and its expression profile in wheat (Triticum aestivum L.) during plant growth and in response to nitrate deficiency

Basketball Action Recognition Algorithm Based on Global Context Awareness

The findings suggest the potential application of the phKL system in wheat chromosome engineering.

Towards Spatial Location Aided Fully-Distributed Dynamic Routing for LEO Satellite Networks
