The antioxidant and antimicrobial properties of two edible mushrooms (Pleurotus ostreatus PBS281009 and Coprinus comatus M8102) were evaluated in this study. The mycelium ethanolic extract obtained in the presence of four different nitrogen sources was investigated with regard to their production of phenolic compounds, beta-carotene, lycopene, and their antimicrobial and antioxidant properties. Significant differences in phenol and beta-carotene concentrations in mycelia grown in different nitrogen sources were observed. Corn extract proved to be the most appropriate source for increasing bioactive properties, leading to the lowest EC50 and minimum inhibitory concentration values in the P. ostreatus PBS281009 species, while peptone was the most suitable for the C. comatus M8102 species. The antimicrobial capacity was screened against Gram-positive and Gram-negative bacteria and against two species of Candida. These results indicate that the mycelia from these two edible mushrooms can be used as nutraceuticals and functional products.

Hexavalent-Chromium-Induced Oxidative Stress and the Protective Role of Antioxidants against Cellular Toxicity

Veer Singh, Nidhi Singh, Manisha Verma, Rashmi Kamal, Ritesh Tiwari, Mahesh Sanjay Chivate, Sachchida Nand Rai, Ashish Kumar, Anupama Singh, Mohan P. Singh, Emanuel Vamanu, Vishal Mishra

Summary: Hexavalent chromium is a highly soluble environmental contaminant that is 100 times more toxic than trivalent chromium. It is mainly derived from leather, chrome plating, coal mining, and paint industries. Hexavalent chromium is recognized as a carcinogen and mutagen, and it can cause damage to multiple organs. Its toxicity depends on its oxidation state, with the reduction to Cr (III) generating reactive oxygen species (ROS) and chromium intermediate species. ROS are responsible for oxidative tissue damage and disruption of cell organelles, and can be neutralized by the antioxidant system.

Dysbiotic Gut Microbiota Modulation by Aronia Fruits Extract Administration

Emanuel Vamanu, Florentina Gatea, Ionela Avram, Gabriel Lucian Radu, Sandeep Kumar Singh

Summary: The administration of chokeberry extract in the GIS1 system was found to have a strong antimicrobial effect and the presence of new organic acids as biomarkers. It gradually increased antioxidant status for up to three months and then decreased along with the reduction of Enterobacteriaceae. Butyric acid was generally unaffected by the antimicrobial effect after three months. The controlled use of this extract corrected the dysbiotic pattern resulting from the long-term use of sweeteners.

Inter and intracellular mitochondrial transfer: Future of mitochondrial transplant therapy in Parkinson?s disease

Rachit Jain, Nusrat Begum, Kamatham Pushpa Tryphena, Shashi Bala Singh, Saurabh Srivastava, Sachchida Nand Rai, Emanuel Vamanu, Dharmendra Kumar Khatri

Summary: Parkinson's disease (PD) is characterized by the degeneration of dopaminergic neurons and the accumulation of alpha-synuclein protein in Lewy bodies, leading to mitochondrial dysfunction. Mitochondrial dynamics, including movement within and between cells, are impaired in PD. Various proteins, enzymes, and molecules are involved in regulating mitochondrial movement. Understanding these pathways is crucial for developing mitochondrial transplant therapy for PD. Techniques such as mitochondrial coculture and mitoception are being explored for their potential translational value. This review focuses on the importance of understanding intercellular and intracellular mitochondrial dynamics in PD and highlights the potential of mitochondrial transplant therapy.

Identification of Prognostic Biomarkers for Suppressing Tumorigenesis and Metastasis of Hepatocellular Carcinoma through Transcriptome Analysis

Divya Mishra, Ashish Mishra, Sachchida Nand Rai, Emanuel Vamanu, Mohan P. Singh

Summary: This study identified 13 hub genes that are overexpressed in both glioblastoma multiforme (GBM) and hepatocellular carcinoma (HCC). It was found that these genes are hypomethylated and contribute to genetic alterations, resulting in chromosomal instability and aneuploidy. A 13-gene predictive model was obtained and validated, suggesting that these hub genes could serve as prognostic biomarkers and potential therapeutic targets for inhibiting tumorigenesis and metastasis.

Demystifying the Role of Prognostic Biomarkers in Breast Cancer through Integrated Transcriptome and Pathway Enrichment Analyses

Divya Mishra, Ashish Mishra, Sachchida Nand Rai, Emanuel Vamanu, Mohan P. Singh

Summary: Breast cancer is a common and deadly disease in women. This study aimed to identify new biomarkers for diagnosis and treatment. Using TCGA datasets, differentially expressed genes (DEGs) were identified and analyzed for functional enrichment and protein-protein interaction networks. Hub genes were selected and their association with overall survival (OS) was analyzed. A total of 824 DEGs were identified, mainly enriched in cell proliferation and signal transduction. Five hub genes, AURKA, BUB1B, CCNA2, CCNB2, and PBK, were found to be associated with poor OS in breast cancer patients. These genes were hypomethylated and involved in chromosomal instability and aneuploidy. The study highlighted the potential of these hub genes as diagnostic and therapeutic targets for breast cancer.

Summary: Metabolic syndrome is a complex condition caused by imbalances in caloric intake and energy expenditure. Plant extracts, with their antioxidant, anti-inflammatory, and insulin-sensitizing properties, offer a promising option for treating metabolic disorders with minimal side effects. However, their limited solubility and stability hinder their effectiveness. The development of green-engineered nanoparticles that enhance the bioavailability and stability of plant-based products has opened up new possibilities for treating metabolic diseases such as obesity, diabetes, and cancer.
