Evaluation of Indigenous Grains from the Peruvian Andean Region for Antidiabetes and Antihypertension Potential Using In Vitro Methods

The health-relevant functionality of 10 thermally processed Peruvian Andean grains (five cereals, three pseudocereals, and two legumes) was evaluated for potential type 2 diabetes-relevant antihyperglycemia and antihypertension activity using in vitro enzyme assays. Inhibition of enzymes relevant for managing early stages of type 2 diabetes such as hyperglycemia-relevant alpha-glucosidase and alpha-amylase and hypertension-relevant angiotensin I-converting enzyme (ACE) were assayed along with the total phenolic content, phenolic profiles, and antioxidant activity based on the 1,1-diphenyl-2-picrylhydrazyl radical assay. Purple corn (Zea mays L.) (cereal) exhibited high free radical scavenging-linked antioxidant activity (77%) and had the highest total phenolic content (8 +/- 1 mg of gallic acid equivalents/g of sample weight) and alpha-glucosidase inhibitory activity (51% at 5 mg of sample weight). The major phenolic compound in this cereal was protocatechuic acid (287 +/- 15 mu g/g of sample weight). Pseudocereals such as Quinoa (Chenopodium quinoa Willd) and Kaniwa (Chenopodium pallidicaule Aellen) were rich in quercetin derivatives (1,131 +/- 56 and 943 +/- 35 mu g [expressed as quercetin aglycone]/g of sample weight, respectively) and had the highest antioxidant activity (86% and 75%, respectively). Andean legumes (Lupinus mutabilis cultivars SLP-1 and H-6) inhibited significantly the hypertension-relevant ACE (52% at 5 mg of sample weight). No alpha-amylase inhibitory activity was found in any of the evaluated Andean grains. This in vitro study indicates the potential of combination of Andean whole grain cereals, pseudocereals, and legumes to develop effective dietary strategies for managing type 2 diabetes and associated hypertension and provides the rationale for animal and clinical studies.

Summary: The study found that Miang extracts fermented by non-filamentous fungi-based and filamentous fungi-based processes had higher phenolic contents and better antioxidant and anti-inflammatory activities compared to young and mature tea leaves. These fermented Miang extracts showed enhanced protection against hydrogen peroxide-induced ROS production in colorectal cells and inhibition of nitric oxide production induced by lipopolysaccharide, indicating their potential as natural bioactive functional ingredients for preventing free radical and inflammatory-mediated diseases.

Summary: This study found that the intake of high-fat, high-carbohydrate (HFHC) meals with orange juice can modulate the expression of miRNAs linked to pancreatic beta-cell function. In a crossover design study involving twelve healthy subjects, it was observed that the intake of HFHC meal + orange juice increased the expression of plasma miR-375, which is a biomarker of pancreatic beta-cell function, and contributed to preventing hyperglycemia.

Bioactive Compounds Intake of the Brazilian Population According to Geographic Region

Summary: Studies have found significant disparities in bioactive compound consumption across populations depending on their geographic origin. The aim of this research was to estimate the intake of bioactive compounds in different regions of Brazil and identify the major contributors. The study analyzed data from the National Dietary Survey 2017-2018, including food intake information from 46,164 individuals aged ≥10 years. The results showed variations in the intake of polyphenols and carotenoids across different regions, with the South having higher polyphenol intake and the Midwest having higher carotenoid intake. Tea and coffee were the main contributors to polyphenol intake in different regions, while acai, caja juice, mango, and corn were important sources of carotenoid intake in specific regions. The findings highlight the potential of food plants in developing marketable products and increasing bioactive compound intake.

Summary: Miang, a traditional fermented tea from Northern Thailand, contains potential probiotic bacteria. Among the 133 lactic acid bacteria (LAB) isolates from Miang, 5 strains were selected for their high tolerance to bile and acidic conditions, inhibitory activity against human pathogens, and survival in simulated gastrointestinal conditions. These strains also showed different responses to antibiotics, cell surface properties, and exhibited antioxidant activity.
