This, along with low sequence identity, suggests convergent evolution. Site-directed mutagenesis experiments were conducted to investigate the functionality of specific residues in DpTPS9.

Summary: In this study, a new macrolide called 25-hydroxy peucemycin was successfully identified from Streptomyces peucetius ATCC 27,952, which exhibited antibacterial effects on certain pathogens. By inactivating a polyketide synthase gene, the biosynthetic gene cluster for the compound was identified, which is considered to be cryptic. The biosynthetic pathway of peucemycin involves five type I polyketide synthase genes encoding eight modules and utilizes unusual extender units.

Summary: In this study, a putative terpene synthase AcTPS3 was identified from Acremonium chrysogenum and confirmed as a sesquiterpene synthase through sequence analysis and in vitro enzyme assay. Metabolic engineered Saccharomyces cerevisiae was constructed to overproduce the product of AcTPS3 for NMR testing. The major product of AcTPS3 was identified as (+)-cubenene, and the optimized S. cerevisiae strain achieved the highest reported biosynthesis titer for (+)-cubenene synthesis.

Summary: Under the guidance of MS/MS-based molecular networking, eight odoriferous sesquiterpenes were isolated from the EtOAc extract of the marine sponge-associated Streptomyces sp. NBU3428. Two undescribed geosmin-type sesquiterpenoid degradations, odoripenoid A and odoripenoid B, and two undescribed germacrane-type sesquiterpenoids, odoripenoid C and odoripenoid D, together with four known related compounds were identified. These compounds displayed potential antifungal activity against Candida albicans.

Summary: Ganoderma sinense, a fungus with a medicinal history of more than 2000 years, is rich in polysaccharides and terpenoids. In this study, researchers identified 21 sesquiterpene synthase genes from G. sinense and found that two of them, GsSTS26 and GsSTS27, are responsible for the synthesis of gleenol, which has insecticidal, antihelminth, and plant growth regulation properties. By modifying E. coli, the researchers were able to significantly increase the production of gleenol, providing a foundation for the industrial production of terpenoids in basidiomycetes.
