MicroRNA-221 and-222 modulate intestinal inflammatory Th17 cell response as negative feedback regulators downstream of interleukin-23

Summary: MiR-221 and miR-222 play a crucial role in regulating intestinal Th17 cell response, with their expression controlled by various cytokines. Loss of miR-221 and miR-222 expression shifts the gene transcription spectrum of Th17 cells to a proinflammatory signature.

Evolving Views of Long Noncoding RNAs and Epigenomic Control of Lymphocyte State and Memory

Tasha A. Morrison, William H. Hudson, Danielle A. Chisolm, Yuka Kanno, Han-Yu Shih, Rafi Ahmed, Jorge Henao-Mejia, Markus Hafner, John J. O'Shea

Summary: Immunological memory is a complex concept with multiple molecular mechanisms. New sequencing technologies have improved the understanding of gene regulation and lymphocyte specification, shedding light on the epigenomic and transcriptomic programs underlying memory. The involvement of long noncoding RNAs (lncRNAs) is increasingly apparent, but their functions and knockout mouse studies are still limited.

A central role for STAT5 in the transcriptional programing of T helper cell metabolism

Alejandro V. Villarino, Arian D. J. Laurence, Fred P. Davisi, Luis Nivelo, Stephen R. Brooks, Hong-Wei Sun, Kan Jiang, Behdad Afzali, Daniela Frasca, Lothar Hennighausen, Yuka Kanno, John J. O'Shea

Summary: This study reveals that STAT5 is a crucial regulator of energy and amino acid metabolism in CD4+ T helper cells, and it plays a role in transcriptional regulation of key enzymes and transporters. Additionally, STAT5 interacts with the mTOR signaling pathway and MYC transcription factor to regulate T-cell metabolism.
