Summary: The study aimed to compare the frequency and phenotype of B, T follicular helper (Tfh), and T follicular regulatory (Tfr) cells in peripheral blood and the cytokine environment present in circulation in children with extended oligoarticular juvenile idiopathic arthritis (extended oligo JIA) and polyarticular JIA (poly JIA) with healthy controls, children with persistent oligo JIA (persistent oligo JIA), and adult JIA patients. The results showed that children with extended oligo JIA and poly JIA had lower frequencies of plasmablasts and regulatory T cells, as well as higher levels of Th17-like Tfh cells in circulation compared to controls. These immunological alterations were not found in adult JIA patients in comparison to controls.

Summary: Rituximab-treated patients and B cell-deficient mice showed reduced expansion of virus-specific CD8(+) T cells after vaccination/infection with different vaccines/pathogens, indicating that B cells can modulate CD8(+) T cell responses. The choice of vaccines for B cell-depleted patients needs to be re-evaluated to effectively induce protective CD8(+) T cell responses.

Summary: This study aimed to investigate the changes in absolute numbers of T-lymphocyte subsets and serum cytokines in patients with systemic sclerosis (SSc). The study found that patients with SSc had lower numbers of Th2 and Treg cells compared to healthy controls, which led to significantly increased ratios of Th1/Th2 and Th17/Treg. Additionally, the levels of certain cytokines were significantly higher in SSc patients. The findings contribute to a better understanding of the pathogenesis of SSc and provide a basis for the development of novel therapeutic interventions targeting these cells and cytokines.

Summary: Activated FLS play a critical role in synovial tissue remodeling and interaction with immune cells in rheumatoid arthritis (RA). Co-culture experiments demonstrated that cytokine-activated FLS enhance T cell activation, proliferation, and differentiation. Transcriptomic analysis revealed a correlation between FLS cytokine signatures and infiltrating CD4(+) T cells in RA synovial tissue. ICAM1 was identified as an important mediator in TNF-mediated FLS-T cell interaction.

Summary: The objective of this study was to investigate the anti-inflammatory and anti-arthritic effects of ellagic acid (EA) in collagen-induced arthritis (CIA). The findings demonstrated that EA significantly reduced inflammation and oxidative stress, and inhibited the development of arthritis in rats. These results suggest that EA could serve as a potential new drug for the treatment of arthritis.

Summary: This study investigated the effects of Fufangmuniziqi formula (FFMN) on Alzheimer's disease (AD) and the role of alkaloids, flavonoids, and saponins in the process. The results showed that FFMN improved cognitive and memory impairment in the AD mouse model. Alkaloids enhanced the cholinergic system, while flavonoids and saponins attenuated neuroinflammation and oxidative stress. These findings suggest that the combination of alkaloid, flavonoid, and saponin fractions derived from FFMN is a promising anti-AD agent.

Summary: This study aimed to explore the hepatoprotective effects of Shicao on CCl4-induced acute liver injury. A rat model of acute liver injury was established and liver function indices were assessed. Untargeted metabolomics was conducted to identify differential metabolites. Network pharmacology was used to construct a network of metabolite-reaction-enzyme-gene constituents, and key components and hub genes were screened. Shicao showed hepatoprotective effects against acute liver injury in rats.
