Summary: This study constructed a single-cell atlas of ESCC and revealed a novel subpopulation of SAA1+ malignant cells that are closely associated with distant metastasis of ESCC. A prediction model based on the gene expression pattern of SAA1+ cells was also established to identify high-risk patients with worse prognosis. These findings enhance our understanding of the mechanisms underlying distant metastasis in ESCC and provide a theoretical basis for improved therapies.

Summary: This study reveals the crucial role of TWIST1 in the establishment of a cancer stem cell-like phenotype, EMT progression, and inhibition of apoptosis in esophageal squamous cell carcinoma (ESCC). These findings provide evidence for TWIST1 as a therapeutic target for suppressing CSC generation and ESCC malignancy.

Summary: In this study, the researchers discovered a signaling interaction between FGF19 and GLI2 that plays a crucial role in the invasion and metastasis of metastatic lung squamous cell carcinoma (LUSC). They found that FGF19 and GLI2 form a positive feedback loop to promote LUSC cell invasion and metastasis, and intervening with this loop may be a strategy for treating FGF19-driven LUSC metastasis.

Summary: p53 mutations predispose cancer cell development, promote their survival and metastasis. No drug that abrogates the oncogenic functions of mutant p53 has been approved for cancer treatment. Whole-genome sequencing of esophageal squamous cell carcinoma (ESCC) tumor tissues and paired normal tissues indicates a higher proportion of TP53 nonsense mutants in our cohort compared to European and American ESCC samples in the IARC database. The most frequent p53 mutations disrupt the inhibition of proliferation, migration, and invasion mediated by wild-type p53 in ESCC.

Summary: This study found that cancer-associated fibroblasts (CAFs) promote metastasis and development of laryngeal squamous cell carcinoma (LSCC) through the ROCK1-mediated epithelial-mesenchymal transition (EMT) via the JAK2/STAT3/ERK1/2 pathway. Inhibition of ROCK1 or JAK2/STAT3/ERK1/2 signal molecules significantly reduces EMT and metastasis.

Summary: PUR alpha promotes cell growth, migration, and invasion in ESCC, likely through inducing EMT to contribute to the progression of ESCC. High expression of PUR alpha is associated with lymph node metastasis, advanced AJCC stage, and worse survival outcomes in ESCC patients.

Summary: Squamous-cell carcinoma is a common type of cancer in humans, with most cases being localized and treatable, but a small percentage of cases that recur or spread can be fatal. Immunocompromised patients are at particularly high risk.

Summary: FSTL1 expression is post-transcriptionally regulated in healthy epithelia and SCC. In healthy cells, binding of KSRP destabilizes FSTL1 mRNA, which is processed as a primary microRNA encoding miR-198. In SCC, downregulation of KSRP terminates miR-198 processing, allowing FSTL1 translation.

Summary: The study found that elevated ATAD2 expression was significantly associated with lymph node metastasis, advanced clinical stage, and poor survival of ESCC patients. ATAD2 promotes ESCC metastasis through regulating the C/EBP beta /TGF-beta 1/Smad3/Snail signaling pathway.

Summary: The aim of this study was to investigate the effect of Mena on TSCC metastasis and its mechanism. The expression of Mena was significantly correlated with lymphatic metastasis and TNM stage, E-cadherin, Vimentin, and MMP2. Mena promotes TSCC invasion and metastasis by inducing the EMT process.

Summary: Transgelin is found to be expressed at low levels in ESCC, and its expression is positively correlated with the prognosis of ESCC patients. It inhibits the malignant progression of ESCC by suppressing the occurrence of EMT, as its overexpression significantly reduces cell invasion and proliferation abilities, while its knockdown promotes cell proliferation, migration, and invasion.

Summary: Recent evidence suggests that cancer stem cells (CSCs) are the origin of cancers. This study investigated the relationship between tumor buds, which are hypothesized to represent epithelial-mesenchymal transition (EMT)-like CSCs, and EMT and CSC markers in cervical squamous cell carcinoma (CxSCC). The findings indicate that tumor buds in CxSCC may represent EMT-like CSCs in vivo.

Summary: The study revealed the pathways and signals in the bulge that control repigmentation in human vitiligo. Through selective capture and RNA-sequencing of cells from the hair follicle bulge, it was found that signals like KCTD10, CTNNB1, and RHO play a role in repigmentation, laying the foundation for the discovery of molecules that induce robust repigmentation in vitiligo.
