Genotype and Tissue-Specific Effects on Alternative Splicing of the Transcription Factor 7-Like 2 Gene in Humans

Context: Noncoding single-nucleotide polymorphisms (SNPs) within the TCF7L2 gene are confirmed risk factors for type 2 diabetes, but the mechanism by which they increase risk is unknown. Objective: We hypothesized that associated SNPs alter TCF7L2 splicing and that splice forms have altered biological roles. Design: Splice forms and 5' and 3' untranslated regions were characterized in sc adipose, muscle, liver, HepG2 cells, pancreas, and islet. Isoform-specific transcript levels were quantified in sc adipose. Alternative splice forms were characterized in HepG2 liver cells under glucose and insulin conditions and in SGBS cells with differentiation. Major isoforms were characterized by transfection. Setting: The study was conducted at an ambulatory general clinical research center. Patients: Patients included 78 healthy, nondiabetic study subjects characterized for insulin sensitivity and secretion. Results: We identified 32 alternatively spliced transcripts and multiple-length 3' untranslated region transcripts in adipose, muscle, islet, and pancreas. Alternative exons 3a, 12, 13, and 13a were observed in all tissues, whereas exon 13b was islet specific. Transcripts retaining exons 13 and 13a but not total TCF7L2 transcripts were significantly correlated with both obesity measures (P < 0.01) and rs7903146 genotype (P < 0.026) in sc adipose. Insulin (5-10 nM) suppressed all TCF7L2 isoforms in SGBS cells but suppressed exon 13a-containing isoforms most significantly (P < 0.001). The isoform distribution differed throughout SGBS cell differentiation. Isoforms with predicted early stop codons yielded stable proteins of the predicted size, bound beta-catenin, and targeted correctly to the nucleus. Conclusions: Intronic TCF7L2 variants may regulate alternative transcript isoforms, which in turn may have distinct physiologic roles. (J Clin Endocrinol Metab 95: 1450-1457, 2010)

ROSEmark: Robust semi-blind ECG watermarking scheme using SWT-DCT framework

This paper proposes a robust semi-blind watermarking scheme, ROSEmark, for authenticating ECG records transmitted through networked devices. Impact of greywater on germination and physiological responses of Triticum aestivum L. HD 2967 in soil amended with poultry biochar

Multi-omic integration reveals cell-type-specific regulatory networks of insulin resistance in distinct ancestry populations

HGSmark: An efficient ECG watermarking scheme using hunger games search and Bayesian regularization BPNN

Summary: The extensive use of telemedicine has increased the exchange of medical data via insecure networks. This paper proposes HGSmark, a watermarking scheme for ECG signals, to address the security and authenticity concerns. The proposed scheme utilizes the HGS algorithm to optimize the embedding strength values and employs a wrapping-based FDCT and SVD for watermarking. Experimental results show that HGSmark achieves high performance in terms of imperceptibility and robustness compared to other state-of-the-art watermarking schemes for ECG signals.

Coswara: A respiratory sounds and symptoms dataset for remote screening of SARS-CoV-2 infection

Two congruent cues are better than one: Impact of ITD-ILD combinations on reaction time for sound lateralization

Summary: This letter presents a reaction time analysis of a sound lateralization test, showing that stimuli from the sides yield quicker reactions and better class accuracy than from the front. Congruent ITD-ILD cues significantly improved both metrics, while opposing ITD-ILD cues resulted in slower responses driven by ITD. These findings corroborate the integrated processing of binaural cues and promote the use of multiple congruent binaural cues in headphone reproduction.

Summary: Using outbred HS rats, expression data, and mediation analysis, we identified QTLs and candidate gene mediators for various metabolic traits. Through physiological and expression QTL mapping, we identified 14 SNP-based pQTLs and 19 haplotype-based pQTLs, with 10 in common. Mediation analysis revealed Grk5, Krtcap3, Ilrun, and Rfx6 as candidate mediators for specific pQTLs. Further verification using gene knockdown/out models confirmed the role of Grk5 and Krtcap3 as regulators of obesity.

Common and ethnic-specific derangements in skeletal muscle transcriptome associated with obesity

Summary: This study compared genes and biological pathways affected by obesity in European and African ancestry populations using data from transcriptome-wide analyses on skeletal muscle tissues. It identified common obesity-associated transcripts and biological pathways across ethnicities, as well as unique obesity-associated processes in either African or European ancestry populations. Integration of SNP-to-Gene analyses predicted 316 critical target genes for obesity-associated genetic variants, suggesting their role in causing obesity.

State Estimation of the Transmission System in the Presence of Unbalanced Load at the Transmission Distribution Boundary

Summary: The conventional transmission system state estimation assumes balanced loads at the transmission distribution boundary, but in reality, distribution feeders have inherently unbalanced loading, especially with the inclusion of small-scale renewable energy sources. Unbalanced loading can cause voltage variation at the boundary and adjacent nodes inside the transmission system. The traditional state estimation method is unable to handle unbalanced nodes. Therefore, this paper proposes a method to detect and identify unbalanced nodes in the transmission system and quickly update their states using a quasi-Newton approach.
