Shaping a Screening File for Maximal. Lead Discovery Efficiency and Effectiveness: Elimination of Molecular Redundancy

High Throughput Screening (HTS) is a successful strategy for finding hits and leads. that have the opportunity to be converted. into drugs. In this paper we highlight novel computational methods used to select compounds to build a new screening file at Pfizer and the analytical, methods we Used to assess their quality. We also introduce the novel concept of molecular redundancy to helps. decide on the density of compounds required in any region: of chemical space in order to be confident of running successful HTS campaigns.

The discovery of a novel series of compounds with single-dose efficacy against juvenile and adult Schistosoma species

Summary: Schistosomiasis, a severe disease caused by parasitic worms, currently relies on praziquantel for treatment, which has limitations such as lack of efficacy against juvenile worms, large dose requirement, and risk of resistance. Researchers have discovered a new series of potent compounds unrelated to praziquantel, with predictions suggesting they could cure infection in humans with a single oral dose 10-fold smaller than praziquantel. Through optimization and design, several compounds in the series have shown efficacy in clearing both juvenile and adult S. mansoni worms in mice with a single oral dose of less than 10 mg/kg, indicating potential for treating schistosomiasis in humans with doses less than 5 mg/kg.
