L-Cysteine and Hydrogen Sulfide Increase PIP3 and AMPK/PPARγ Expression and Decrease ROS and Vascular Inflammation Markers in High Glucose Treated Human U937 Monocytes

Diabetic patients have lower blood levels of L-cysteine (LC) and hydrogen sulfide (H2S) and a higher incidence of vascular inflammation. This study examined whether impaired LC or H2S levels affect vascular inflammation markers in diabetes. Human U937 monocytic cells were treated with high-glucose (HG, 25mM, 20h) in the presence or absence of LC (100, 500, or 1,000 mu M, an endogenous precursor of H2S) or Na2S (5 or 25 mu M, an exogenous source of H2S). Both LC and Na2S supplementation decreased intracellular ROS production and increased cellular PIP3 (phosphatidylinositol-3,4,5-trisphosphate) in HG-exposed cells. The effect of LC on PIP3 was prevented by propargylglycine, an inhibitor of cystathionine--lyase (CSE) that catalyzes H2S formation from LC. Signal silencing studies with CSE siRNA also showed the inhibition of H2S formation and PIP3 upregulation in LC-supplemented CSE knockdown cells exposed to HG. This demonstrates that H2S plays a role in mediating the effect of LC on increased PIP3. Using the PI3K specific inhibitor LY294002, this study demonstrated that PI3K activation mediates the effect of LC and Na2S on PIP3 upregulation. Results showed that supplementation with LC and Na2S reduced NF-B phosphorylation and the secretion of TNF-, MCP-1, IL-8, IL-1, and IP-10. Treatment with LC (500 mu M), Na2S (25 mu M), and PIP3 (5nM) increased the AMPK phosphorylation and PPAR expression in cells exposed to HG. This study reports for the first time a novel molecular mechanism by which Na2S or LC supplementation can lower oxidative stress and various markers of vascular inflammation in diabetes. J. Cell. Antidiabetic potential of soy protein/peptide: A therapeutic insight

Summary: Data science has played a crucial role in the COVID-19 pandemic response, and this study demonstrates how a statistical and machine learning approach can be used to discriminate between SARS-CoV-2 infection and immune response to a whole virion vaccine. By analyzing serial antibody data and incorporating an ensemble ML model, the researchers were able to accurately classify potential infections and validate their findings through neutralization assays. This study highlights the importance of real-world vaccine effectiveness assessments for whole virion vaccines.

Summary: This study showed that Premna herbacea and its active constituent, Verbascoside, have a beneficial effect on hepatic steatosis caused by a high fat diet. By regulating the AMPK/SREBP/ACC/HMGCR signaling pathways, Premna herbacea and Verbascoside can prevent and improve hepatic steatosis. This suggests that Premna herbacea may be a potential functional food for managing hepatic steatosis.

Hydrogen Sulfide Regulates Irisin and Glucose Metabolism in Myotubes and Muscle of HFD-Fed Diabetic Mice

Summary: Irisin, a novel myokine, is involved in glucose homeostasis through the activation of PGC-1 alpha/FNDC5/Irisin signaling pathway. This study found that deficiency of hydrogen sulfide (H2S) leads to decreased secretion of FNDC5 and Irisin and alters glucose metabolism, while upregulation of H2S levels has beneficial effects on glucose homeostasis.

Summary: This study examined the antidiabetic efficacy of popular fermented soybean foods from Northeast India. Results showed that aqueous extract of Hawaijar significantly augmented glucose utilization and improved glucose metabolism in cultured myotubes and diabetic rats. Chemical profiling suggests the possible role of bioactive proteins/peptides and isoflavones underlying the antihyperglycemic potential of Hawaijar. These findings may contribute to the development of food-based prophylactics/therapeutics for managing hyperglycemia.

The Positive Association of Plasma Levels of Vitamin C and Inverse Association of VCAM-1 and Total Adiponectin with Bone Mineral Density in Subjects with Diabetes

Summary: Mesoporous silica nanoparticles (MSNs) are extensively used in biomedical applications such as drug delivery, diagnosis, bio-sensing, and cell tracking due to their unique properties. MSNs have been extensively studied in drug delivery field due to their highly ordered mesopores, high surface area, stability, and biocompatibility. MSN-based nanocomposites have been utilized for delivering therapeutic molecules for diabetes management. This review focuses on the delivery of antidiabetic therapeutics with emphasis on the functionalization of MSNs and stimuli-responsive delivery.

Porous Nitrogen-Doped Crumpled Graphene Nanoparticles: A Metal-Free Nanozyme for Selective Detection of Dopamine in Aqueous Medium and Human Serum

Summary: This article describes the synthesis of porous 3D nitrogen-doped crumpled graphene nanoparticles (CGNPs) and their use as a platform for the sensitive detection of dopamine (DA) in complex biological media such as blood serum. The CGNPs were prepared by doping graphene oxide (GO) using ammonium hydroxide in a hydrothermal treatment. The system selectively detected DA with a limit of detection of 1.15 μM and a linearity range of 1 to 20 μM, and it showed good selectivity in the presence of various interfering species and in human blood serum.

Summary: The incidence of Alzheimer's disease is higher in people over the age of 65 and in African Americans. This study discovered a negative association between plasma AChE levels and H2S levels, and a positive association between AChE levels and age, NLR, ROS, and HbA1c levels in African Americans. The study also suggests that hyperglycemia and lower H2S levels may contribute to an increase in AChE activity.
