Polarized cells such as epithelial cells and neurons exhibit different plasma membrane domains with distinct protein compositions. Recent studies have shown that sorting of transmembrane proteins to the basolateral domain of epithelial cells and the somatodendritic domain of neurons is mediated by recognition of signals in the cytosolic domains of the proteins by adaptors. These adaptors are components of protein coats associated with the trans-Golgi network and/or recycling endosomes. The clathrin-associated adaptor protein 1 (AP-1) complex plays a preeminent role in this process, although other adaptors and coat proteins, such as AP-4, ARH, Numb, exomer, and retromer, have also been implicated.

The autophagy protein ATG9A enables lipid mobilization from lipid droplets

Summary: ATG9A is a transmembrane protein involved in lipid mobilization, essential for autophagosome expansion, mitochondrial fatty acid import, and beta-oxidation. Depletion of ATG9A inhibits autophagy, increases lipid droplet size/number, and blocks transfer of fatty acids to mitochondria. ATG9A localizes to vesicular-tubular clusters associated with TMEM41B and plays a critical role in lipid mobilization from lipid droplets to autophagosomes and mitochondria.

Summary: This study investigates the processing and sorting of NRG3 in neurons. It finds that NRG3 is cleaved in the trans-Golgi network and released, and then re-endocytosed and transported into axons. NRG3 accumulates at presynaptic terminals through stable interaction with its receptor. This trans-synaptic retention mechanism may explain the polarized expression of other neuronal transmembrane ligands and receptors.

The results showed that Rab2a and Rab7 are present in late endosomes, while Arl8 and BORC are present in lysosomes, and HOPS drives LE-LYS fusion by bridging late endosomal Rab2a with lysosomal BORC-anchored Arl8. Additionally, Rab7 promotes fusion by moving LEs towards LYSs via dynein.

Inhibition of endolysosome fusion increases exosome secretion

Summary: This study shows that disruption of the BORC-ARL8-HOPS pathway increases exosome secretion by impairing fusion of multivesicular endosomes with lysosomes. This leads to the release of intraluminal vesicles into the extracellular space. Therefore, suppressing the BORC-ARL8-HOPS pathway could be used to enhance exosome yields.

Intrathecal AAV9/AP4M1 gene therapy for hereditary spastic paraplegia 50 shows safety and efficacy in preclinical studies

Summary: Spastic paraplegia 50 (SPG50) is an ultrarare childhood-onset neurological disorder caused by biallelic loss-of-function variants in the AP4M1 gene. In preclinical studies, adeno-associated virus (AAV)/AP4M1 gene therapy showed promising results in rescuing the symptoms of SPG50. The therapy was effective in transducing patient-derived fibroblasts and demonstrated dose-dependent effects in Ap4m1-KO mice, supporting the potential for a clinical trial in treating SPG50 through intrathecal administration of AAV9/AP4M1.
