Summary: Axon remodeling through sprouting and pruning is important for the refinement of developing neural circuits. Developing sensory axons deprived of neurotrophic support undergo apoptosis, with the tumor suppressor P53 playing a crucial role in activating the apoptotic pathway in axons. P53 appears to act in the cytoplasm to directly activate the mitochondrial apoptotic pathway in axons, highlighting a cytoplasmic role in the anterograde death of developing sensory axons.

Summary: Combining FLT3 TKIs with BCL-2 selective inhibitors synergistically reduces cell proliferation and enhances apoptosis, resensitizing FLT3 TKI-resistant cells. Venetoclax mitigates the pro-survival effects of FLT3 TKIs by dissociating BIM from BCL-2 and decreasing BIM expression. This study provides evidence that adding BCL-2 inhibitors enhances the efficacy of FLT3 TKI therapy in FLT3/ITD AML treatment.

It identifies methylation of a regulatory CpG island within the PUMA promoter as a mechanism for mediating PUMA downregulation and resistance to venetoclax. The loss of PUMA results in metabolic reprogramming and higher oxidative phosphorylation, resembling the metabolic phenotype associated with venetoclax resistance. Additionally, it highlights the critical role of BAX-mediated apoptosis in drug resistance and provides potential therapeutic targets to overcome venetoclax resistance.

In this article, we summarize the research on SUP orthologs in plant species other than Arabidopsis, with a focus on the legume Medicago truncatula. The MtSUP gene in M. truncatula is involved in the complex genetic network controlling compound inflorescence and flower development in legumes, providing new insights into these processes.
