Summary: The study investigated the therapeutic benefits of adipose tissue derived-mesenchymal stem cells (ASCs) and ASC-conditioned medium (CM) in promoting myelination and axonal regeneration. Results showed that both ASCs and CM could potentially aid in nerve regeneration, especially in reversing the effects of TNF-induced nerve damage on myelination-related genes and proteins.

Summary: This study investigated the role of syndecan-1 and heat shock protein 70 (HSP-70) in mediating the regenerative effects of adipose-derived mesenchymal stem cells (ADSCs) on peripheral nerves. The results showed that ADSCs could secrete syndecan-1 and HSP-70 in response to stress conditions, and ADSC treatment significantly improved functional recovery and nerve regeneration.

Summary: Autologous fat grafting is commonly used in reconstructive surgery to improve functional form. Fat tissue enrichment with adipose-derived stem cells can increase the long-term stability of grafts. Basic science research suggests that adipose-derived stem cells can enhance nerve regeneration and revascularization, and reduce nerve fibrosis. Clinical studies show promising results in secondary carpal tunnel release revisions using autologous lipofilling.

Summary: Topically applied exosomes from adipose-derived stem cells and FK506-pretreated adipose-derived stem cells significantly enhanced nerve regeneration after injury, with histone deacetylases, amyloid-beta A4 protein, and integrin beta-1 identified as potential proteins involved in this process.

Summary: An injectable decellularised matrix hydrogel (DAM-gel) was developed to simulate the stem cell microenvironment, promoting the repair of peripheral nerve defects with adipose-derived mesenchymal stem cells (ADSCs). The ADSC-loaded DAM-gel significantly improved Schwann cell proliferation, axonal regeneration, and neurological function recovery in a rat model. These findings suggest that DAM-gels have great potential for enhancing the repair capacity of ADSCs in peripheral nerve defects.

Summary: This article reviews emerging strategies to improve the therapeutic efficacy of adipose-derived stem cells (ADSCs) in tissue regeneration. These strategies include advanced cell expansion technologies, enhancing regenerative functions of ADSCs, and improving cell delivery efficiency. Critical issues about the sources, effectiveness, and safety of ADSCs are also pointed out.

Summary: The study investigated three differentiation protocols for differentiating rat ASCs into specialized SC phenotypes, comparing their expressions of neurotrophins and myelin markers. The results showed diverse expression profiles of differentiated ASCs, with different protocols inducing varying levels of expression of neurotrophins and myelin markers.

Summary: Our study demonstrates that a diabetic environment has an adverse effect on ASCs, and adopting a low glucose culture condition is a simple and effective way to enhance the wound healing capabilities of diabetic ASCs. This finding is valuable for the clinical application of autologous ASCs from diabetic patients.

Summary: The research found that graphene oxide is a suitable substrate for human sAD-MSC incubation, promoting regeneration of severe peripheral nerve injuries. However, transcriptome changes are minimal, and growth factor gene expression remains unchanged on GO substrates, but NGF and GDNF protein secretion increases at day 3 and 7. In general, GO substrates are compatible with sAD-MSCs primed for peripheral nerve regeneration strategies and protect the cell population in harsh environments.

Summary: The behavior of nerve cells is crucial in nerve regeneration, with the surrounding mechanical, topographical, and electrical microenvironment playing a key role in influencing nerve cell behavior. Recently, biological scaffolds have been developed with various physical properties to regulate nerve cell behavior conversion, providing a strong driving force for nerve regeneration. Research has focused on the biological basis of mechanical transduction in nerve cells, as well as the impact of physical cues on neurite outgrowth and neural stem cell differentiation.

Summary: Opioids have been the mainstay for postoperative pain management in brain surgery, but their use is limited due to potential side effects; multimodal analgesia targets pain pathways with different mechanisms of actions, resulting in additive and/or synergistic effects to provide adequate pain relief while minimizing drug-induced side effects.
