The Protease Allergen Pen c 13 Induces Allergic Airway Inflammation and Changes in Epithelial Barrier Integrity and Function in a Murine Model

Fungal allergens are associated with the development of asthma, and some have been characterized as proteases. Here, we established an animal model of allergic airway inflammation in response to continuous exposure to proteolytically active Pen c 13, a major allergen secreted by Penicillium citrinum. In functional analyses, Pen c 13 exposure led to increased airway hyperresponsiveness, significant inflammatory cell infiltration, mucus overproduction, and collagen deposition in the lung, dramatically elevated serum levels of total IgE and Pen c 13-specific IgE and IgG1, and increased production of the Th2 cytokines IL-4, IL-5, and IL-13 by splenocytes stimulated in vitro with Pen c 13. To examine the mechanisms involved in the regulation of allergenicity by Pen c 13, we performed two-dimensional fluorescence difference gel electrophoresis analysis combined with nano-LC-MS/MS, followed by bioinformatics analysis to identify potential targets that associated with allergic inflammation, which suggested that galectin-3 and laminin might be involved in novel pathogenic mechanisms. Finally, we focused on junctional proteins between cells, because, in addition to opening of the epithelial barrier by environmental proteases possibly being the initial step in the development of asthma, these proteins are also associated with actin rearrangement. Taken together, our findings indicate that Pen c 13 exposure causes junctional structure alterations and actin cytoskeletal rearrangements, resulting in increased permeability and airway structural changes. These effects probably change the lung microenvironment and foster the development of allergic sensitization.

Topical Melatonin Exerts Immunomodulatory Effect and Improves Dermatitis Severity in a Mouse Model of Atopic Dermatitis

Summary: Oral melatonin supplement has been shown to improve dermatitis severity in children with AD. This study investigated the mechanism and immunomodulatory effect of topical melatonin treatment in mice and cell models of AD. The results suggest that melatonin may have a direct immunomodulatory effect on AD and could be a potential treatment for patients with AD.

Predictive characteristics to discriminate the longitudinal outcomes of childhood asthma: a retrospective program-based study

It finds that blood eosinophil percentage and mold sensitization are significantly associated with increased asthma severity. Characteristics including sensitization status and ICS dosage demonstrate an improved capability for predicting clinical outcomes.

Circulating SSEA-1+ stem cell-mediated tissue repair in allergic airway inflammation

Summary: Endogenous adult lung SSEA-1(+) cells almost disappear in asthmatic mice, but a unique population of circulating SSEA-1(+) cells is enriched after the challenge phase. Clinical features and outcomes of patients with chronic granulomatous disease in Taiwan

Summary: This study aimed to investigate the clinical features, genetic characteristics, treatment, and prognosis of patients with chronic granulomatous disease (CGD). Results showed that there were six patients with X-linked CGD and three patients with autosomal recessive CGD. Patients with X-linked CGD had earlier onset and diagnosis ages compared to those with autosomal recessive CGD. Prophylactic use of antibiotics, antifungal agents, and interferon-gamma might alleviate infection episodes in CGD patients. Rating: 7 out of 10.

Characterization of the biomarkers related to the clinical course and outcomes of juvenile dermatomyositis

Summary: This study aimed to evaluate the clinical characteristics of children diagnosed with juvenile dermatomyositis (JDM) in Taiwan and identify biomarkers for predicting the disease course and outcomes. The study included 47 patients, with a mean age of 7.5 years at disease onset. The most common initial presentations were Gottron's sign, muscle weakness, and facial rash. The results identified factors associated with complete clinical remission and the chronic continuous course, highlighting the importance of aggressive treatment for the latter.

M2-like macrophages polarized by Foxp3- Treg-of-B cells ameliorate imiquimod-induced psoriasis

Summary: Our study demonstrated that splenic B cells can convert CD4(+)CD25(-) naive T cells into CD4(+)CD25(+)Foxp3(-) regulatory T cells without adding cytokines, termed Treg-of-B cells. These Treg-of-B cells are potent suppressors of adaptive immunity. We investigated whether Treg-of-B cells could promote M2 macrophage polarization and alleviate psoriasis. Our results showed that Treg-of-B cells can induce M2 macrophage polarization and attenuate the clinical manifestations of psoriasis.

Risk factors for subsequent lupus nephritis in patients with juvenile-onset systemic lupus erythematosus: a retrospective cohort study

Summary: This study aimed to determine the risk factors for subsequent lupus nephritis (LN) development in patients with juvenile-onset systemic lupus erythematosus (jSLE). A retrospective cohort study found that out of 48 jSLE patients without initial LN, 20 developed subsequent LN. Persistently high levels of anti-double-stranded DNA antibody and erythrocyte sedimentation rate were identified as risk factors for subsequent LN in jSLE patients.
