Stem-like Cancer Cells Are Inducible by Increasing Genomic Instability in Cancer Cells

The existence of cancer stem cells (CSCs) or stem-like cancer cells (SLCCs) is regarded as the cause of tumor formation and recurrence. However, the origin of such cells remains controversial with two competing hypotheses: CSCs are either transformed from tissue adult stem cells or dedifferentiated from transformed progenitor cells. Compelling evidence has determined the chromosomal aneuploidy to be one of the hallmarks of cancer cells, indicating genome instability plays an important role in tumorigenesis, for which CSCs are believed to be the initiator. To gain direct evidence that genomic instability is involved in the induction of SLCCs, we utilized multiple approaches to enhance genomic instability and monitored the percentage of SLCC in cultured cancer cells. Using side population (SP) cells as a marker for SLCC in human nasopharyngeal carcinoma (NPC) and CD133 for human neuroblastoma cells, we found that DNA damage inducers, UV and mitomycin C were capable of increasing SP cells in NPC CNE-2 and neuroblastoma SKN-SH cells. Likewise, either overexpression of a key regulator of cell cycle, Mad2, or knock down of Aurora B, an important kinase in mitosis, or Cdh1, a key E3 ligase in cell cycle, resulted in a significant increase of SP cells in CNE-2. More interestingly, enrichment of SP cells was observed in recurrent tumor tissues as compared with the primary tumor in the same NPC patients. Our study thus suggested that, beside transformation of tissue stem cells leading to CSC generation, genomic instability could be another potential mechanism resulting in SLCC formation, especially at tumor recurrence stage.

Downregulation of HINFP induces senescence-associated secretory phenotype to promote metastasis in a non-cell-autonomous manner in bladder cancer

Summary: The downregulation of histone H4 transcription factor (HINFP) is associated with cell senescence in bladder cancer tissues, and lower HINFP expression predicts an unfavorable outcome in bladder cancer patients. Knockout of HINFP inhibits the transcription of H1F0 and H1FX, leading to DNA damage and cell senescence, which suppresses the proliferation and growth of bladder cancer cells. However, the senescence-associated secretory phenotype enhances the invasion and metastasis of non-senescent bladder cancer cells. Histone deacetylase inhibitors can effectively eliminate the senescent cells induced by HINFP knockout, thereby suppressing the invasion and metastasis of bladder cancer cells.

Targeting the Lysosomal Degradation of Rab22a-NeoF1 Fusion Protein for Osteosarcoma Lung Metastasis

Summary: The Rab22a-NeoF1 fusion protein undergoes lysosomal degradation mediated by the autophagy receptor NDP52 and the E3 ligase STUB1. The degradation is facilitated by the phosphorylation of Rab22a-NeoF1 by PINK1 kinase. Sorafenib and Regorafenib can inhibit osteosarcoma lung metastasis induced by Rab22a-NeoF1 by upregulating PINK1. These findings suggest that targeting the lysosomal degradation of the Rab22a-NeoF1 fusion protein may be beneficial for osteosarcoma patients.

Intercellular transfer of activated STING triggered by RAB22A-mediated non-canonical autophagy promotes antitumor immunity

Summary: Activated STING can be transferred between cells to promote antitumor immunity, a process triggered by non-canonical autophagy mediated by RAB22A. RAB22A induces the fusion of non-canonical autophagosomes with early endosomes, forming a new organelle called Rafeesome, and suppresses fusion with lysosomes. The inner vesicle of non-canonical autophagosome, containing activated STING, is secreted as R-EV, which induces the release of IFN beta from recipient cells and promotes antitumor immunity.
