Summary: A truncated mutant OUC-FaKC16Q was constructed and found to have hydrolytic activity on carrageenan oligosaccharides. This research expands the product spectrum of OUC-FaKC16A and highlights its potential in the biomanufacturing of carrageenan oligosaccharides.

Summary: In this study, x-carrageenase was successfully immobilized, and the immobilized enzyme demonstrated better stability and storage stability compared to the free enzyme. The produced KCOs by the immobilized enzyme were able to decrease lipid levels and activate AMPK activity, suggesting their potential as a therapeutic agent for preventing hyperlipidemia.

Summary: In this study, a C-terminal bacterial immunoglobulin-like domain (Big_2) was identified in kappa-carrageenase and found to be essential for maintaining the activity and thermostability of the enzyme. The noncatalytic Big_2 domain was also shown to contribute to the processivity of the enzyme and could enhance hydrolysis of kappa-carrageenan independently. Additionally, sequence analysis revealed the conservation of Big_2 domains in bacterial kappa-carrageenases, suggesting their universal noncatalytic functions.

Summary: Comparative structural analysis was carried out on gelling polysaccharides from A. flabelliformis and M. pacificus, and their effect on human colon cancer cell lines was studied. The polysaccharides from M. pacificus predominantly contained kappa units, while the polysaccharides from A. flabelliformis predominantly contained iota units. Oligosaccharides derived from these polysaccharides showed antiproliferative effects on the tested cell lines, with kappa/iota-oligosaccharides having a greater effect on colony formation and iota/kappa-oligosaccharides having a stronger inhibitory effect on cell migration.
