Summary: In this study, the bacterial glycosyltransferase YjiC1 was used for glycosylation of triterpenoids from Antrodia camphorata. Eleven new compounds were obtained through enzymatic reactions. Glucosylation enhanced the inhibitory activities against COX-2 and improved the anti-inflammatory activities of Antrodia ergostanes on acute lung injury mice, especially (25R)-antcin C 7-O-beta-d-glucoside.

Summary: Eleven new compounds were obtained from enzymatic reactions using the bacterial glycosyltransferase YjiC1 to glycosylate triterpenoids from the medicinal fungus Antrodia camphorata. Glucosylation could increase the inhibitory activities against COX-2, and improve the anti-inflammatory activities of Antrodia ergostanes on acute lung injury mice, particularly (25R)-antcin C 7-O-beta-d-glucoside.

It was found through network pharmacology analysis that MAPK3 was the major action target of Antcin A. Experimental research indicated that Antcin A suppressed mouse liver injury, reduced inflammatory factor levels, and enhanced anti-oxidative capacity. It also suppressed the expression of MAPK3 and the downstream NF-KB signal.

Summary: Non-alcoholic fatty liver disease (NAFLD) is a common health issue. This study investigated the beneficial effect of Typhaneoside (TYP) on cells and mice with disorders of glucose and lipid metabolism, inflammation, oxidative stress and lower thermogenesis, mediated through the activation of the farnesoid X receptor (FXR). TYP improved metabolic parameters, such as blood glucose, lipid accumulation, insulin resistance, inflammation, oxidative stress and energy expenditure in vitro and in vivo by activating the expression of FXR.

Summary: This study found that adhesion G protein-coupled receptor F1 (Adgrf1) regulates hepatic lipid metabolism by controlling the expression of stearoyl-coA desaturase 1 (Scd1). The downregulation of Adgrf1 expression in the livers of obese subjects may serve as a protective mechanism to prevent the excessive accumulation of fat. These findings not only reveal a new mechanism for the progression of non-alcoholic fatty liver disease (NAFLD), but also propose a novel therapeutic approach to combat NAFLD by targeting Adgrf1.

Summary: In recent years, metabolomics techniques have identified many specific metabolites related to nonalcoholic fatty liver disease (NAFLD). This study aimed to examine the candidate targets and molecular pathways involved in NAFLD when iron overload is present. Male Sprague Dawley rats were fed control or high-fat diets with or without excess iron, and metabolomics analysis was performed using UPLC-MS. The results showed that a high-fat, high-iron diet resulted in increased triglyceride accumulation and oxidative damage, and several metabolites were significantly altered in the presence of iron overload.

Summary: This study investigated the protective effect of Antrodin C, a fermented metabolite of Antrodia camphorata, on liver fibrosis. The results showed that Antrodin C inhibited the activation, migration, and extracellular matrix accumulation of hepatic stellate cells, and reduced the degree of CCl4-induced liver fibrosis. Antrodin C also improved the abnormal levels of liver enzymes in serum. Mechanistically, Antrodin C exerted its anti-fibrotic activity through modulation of several signaling pathways. Overall, Antrodin C could serve as a protective molecule against liver fibrosis.
