Polyphenolic composition of grape stem extracts affects antioxidant activity in endothelial and muscle cells

The aim of the present study was the assessment of the antioxidant effects of polyphenolic extracts derived from the stems of three Greek grape varieties (Moshomayro, Mavrotragano and Mandilaria) in endothelial (EA.hy926) and muscle (C2C12) cells. We also investigated the effects of the polyphenolic composition on the antioxidant effects of the grape stem extracts. For this purpose, the endothelial and muscle cells were treated with low non-cytotoxic concentrations of the extracts for 24 h in order to assess the effects of the extracts on cellular redox status using oxidative stress biomarkers. The oxidative stress markers were thiobarbituric acid reactive substances (TBARS), protein carbonyl (CARB) levels, reactive oxygen species (ROS) levels and glutathione (GSH) levels. The results revealed that treatment of the EA.hy926 cells with Mandilaria extract significantly decreased the TBARS levels by 14.8% and the CARB levels by 25.9 %, while it increased the GSH levels by 15.8% compared to the controls. Moreover, treatment of the EA.hy926 cells with Mavrotragano extract significantly increased the GSH levels by 20.2%, while it significantly decreased the TBARS and CARB levels by 12.5% and 16.6%, respectively. Treatment of the C2C12 cells with Mandilaria extract significantly decreased the TBARS levels by 47.3 %, the CARB levels by 39.0 % and the ROS levels by 21.8%, while it increased the GSH levels by 22.6% compared to the controls. Moreover, treatment of the C2C12 cells with Mavrotragano significantly decreased the TBARS, CARB and ROS levels by 36.2%, 35.9% and 16.5%, respectively. In conclusion, to the best of our knowledgel, our results demonstrate for the first time that treatment with grape stem extracts at low concentrations improves the redox status of endothelial and muscle cells. Thus, grape stem extracts may be used for developing antioxidant food supplements or biofunctional foods. However, it was also found that the polyphenolic composition of grape stem extracts affects their antioxidant capacity. For example, the results suggested that trans-resveratrol, gallic acid, (+)-catechin, ferulic acid, caffeic acid, quercetin, coumaric acid and kaempferol may be essential for the antioxidant activity of grape stem extracts.

Renoprotective Effect of Taxifolin in Paracetamol-Induced Nephrotoxicity: Emerging Evidence from an Animal Model

Summary: This study aimed to evaluate the renoprotective effects of taxifolin (TXF) in a paracetamol (PAR)-induced nephrotoxicity rat model. The results showed that the levels of blood urea nitrogen, creatinine and malondialdehyde were significantly increased in the PAR group, while the levels of glutathione peroxidase, glutathione reductase and total glutathione were significantly higher in the TXF group. Histopathological examination revealed severe tubular epithelium damage in the PAR group, while the TXF group showed mild dilated/congested blood vessels. These findings suggest that TXF has potential renoprotective effects against PAR-induced nephrotoxicity through its antioxidant activity and should be further investigated.

Species composition and ecological structure of ground beetles (Coleoptera, Carabidae) communities as biological indicators of the agro-environmental sustainability

Summary: The detrimental environmental consequences caused by intensification of crop cultivation can be prevented through monitoring of specific biological indicators. This study focused on the impact of crop type and cultivation intensity on the community of ground beetles in the forest-steppe of Western Siberia. The ground beetles' community showed a high evenness of species distribution across the agroecosystems. Differences in ground beetle distribution were observed between wheat and corn crops. The use of bioindicators allowed for assessing the environmental sustainability of the agricultural environment and provided insights for ecologically directed correction of agrotechnological operations in agroecosystem management.

Summary: The aim of our study was to evaluate the differences among different groups of drug users, focusing on hematological, biochemical, urinary, and cardiological aspects. A retrospective study was conducted on 2572 hospitalized patients in Bucharest, Romania from 2015 to 2021. The study group was characterized based on demographic data, personal history/co-morbidities, consumption history, and substitution treatment. The analysis of medical records revealed correlations between GGT levels and alcohol intake as well as contagious diseases, a decrease in MCH and MCHC levels across the entire study group, and correlations between specific cardiological events and certain substance groups. The study provides important information on different types of drug users and supports future research on the chemical structures of consumed compounds.

Role of gut microbiota in the modulation of the health effects of advanced glycation end-products (Review)

Summary: The aim of this study was to summarize the potential interactive effects between the gut microbiota and advanced glycation end-product (AGE) accumulation and toxicity in the host, and to reveal the mediating effects of the gut microbiota on AGE-related health effects. Existing data demonstrates that dietary AGEs can significantly impact the richness and diversity of the gut microbiota, with specific effects depending on species type and exposure dose. Additionally, the gut microbiota may metabolize dietary AGEs. The characteristics of the gut microbiota, including richness and relative abundance of certain taxa, are closely associated with AGE accumulation in the host organism. In turn, the interplay between AGE toxicity and modulation of the gut microbiota may contribute to aging and diabetes-associated diseases. Bacterial endotoxin lipopolysaccharide appears to mediate the interactions between the gut microbiota and AGE toxicity, specifically through modulation of the receptor for AGE signaling. Therefore, modulation of the gut microbiota using probiotics or other dietary interventions may significantly impact AGE-induced glycative stress and systemic inflammation.

DNA Damage Estimation after Chronic and Combined Exposure to Endocrine Disruptors: An In Vivo Real-Life Risk Simulation Approach

Summary: Exposure to combined endocrine disruptors can cause DNA damage, as observed in this study using comet and micronuclei assays. The highest level of damage was found in the group exposed to a high-dose mixture of substances, particularly glyphosate. The micronuclei assay results were moderately correlated with exposure period, and all exposure groups showed an increasing trend in micronuclei counts over time.

Summary: Cancer is a major issue in clinical, social, and economic aspects, ranking highest in terms of cause-specific disability-adjusted life years among all human pathologies. The trigger of cancer involves various factors, including genetic predisposition and telomere status, which are DNA structures at the end of chromosomes. However, there is no universal trend in the association between telomere length and cancer, as both short and long telomeres have been linked to high cancer incidence risks. This review aims to present the complex relationship between telomere length and cancer comprehensively.

Redox Biomarkers Assessment after Oral Administration of Wine Extract and Grape Stem Extract in Rats and Mice

Summary: Wine and its by-products, such as grape stems, contain bioactive polyphenolic compounds that can potentially benefit animal and human health. However, there are limited studies on the safety and toxicological risks of these compounds in vivo. This study examined the effects of oral administration of grape stem and wine extracts on redox biomarkers in mice and rats. The results showed that grape stem extract improved redox homeostasis in mice, while wine extract had both beneficial and harmful effects on rat redox status, indicating the complexity of biological systems.

Summary: Human milk is the healthiest option for newborns, but infant formula is a valuable alternative in certain circumstances. Ensuring pollutant-free composition, infant formulas are regulated and monitored for safe exposure. While legislation varies across countries, standard policies for protecting vulnerable infants are in place. This article presents current regulations and directives for limiting endocrine-disrupting chemicals and persistent organic pollutants in infant formulas, highlighting the need for limited risk assessment studies to assess the health risks for infants from dietary exposure to pollutants.

Summary: This study aimed to investigate the impact of carnosic acid (CA) on indomethacin (IND)-induced gastric ulceration. The results showed that CA pretreatment attenuated both gross morphological lesions and histopathological alterations induced by IND, and it achieved gastroprotective effects by reducing oxidative stress and suppressing inflammation.

Chlorogenic Acid Attenuates Doxorubicin-Induced Oxidative Stress and Markers of Apoptosis in Cardiomyocytes via Nrf2/HO-1 and Dityrosine Signaling

Summary: This research evaluated the cardioprotective properties of chlorogenic acid (CGA) in doxorubicin (DOX)-induced cardiotoxicity and identified one of the possible mechanisms for its protective effects. The study found that CGA ameliorated oxidative stress and cardiomyocyte apoptosis by upregulating the Nrf2/HO-1-dependent pathway and downregulating dityrosine (DT) expression. These findings suggest that CGA may be cardioprotective, particularly in patients receiving DOX-based chemotherapy.

Reproductive Effects of S. boulardii on Sub-Chronic Acetamiprid and Imidacloprid Toxicity in Male Rats

Summary: The study aimed to investigate the positive effects of probiotic Saccharomyces boulardii on the male reproductive system under low dose neonicotinoid pesticide exposure conditions. Acetamiprid and imidacloprid caused degeneration and necrosis of spermatocytes, severe edema of the intertubular region, and hyperemia. This was accompanied by increased oxidative stress and apoptosis. Saccharomyces boulardii supplementation mitigated these toxic effects.

Summary: The wide-scale emergence of glyphosate-resistant weeds has caused an increase in the application of herbicide mixtures containing glyphosate plus dicamba or glyphosate plus 2,4-D. This raises concerns about the environmental and health risks from exposure to a mixture of herbicide active ingredients. The study evaluated the hepatotoxic effects of perinatal exposure to glyphosate alone or in combination with 2,4-D and dicamba on Wistar rats. The findings highlight the importance of assessing the combined effects of chemical pollutants at safe individual doses.

Summary: This study found that chronic diseases of the urogenital tract can develop under the effects of chemical hazards in the working environment, particularly from nanosized particles generated in industrial processes. The researchers studied the nephrotoxicity of element oxide nanoparticles through animal experiments, and found that these nanoparticles had a toxic effect on the kidneys and caused changes in kidney function and histological structure. The results of this study are important for the early diagnosis of occupational and nonoccupational nephropathies.
