Summary: This article presents a new method called Backbone-Installed Split Intein-Assisted Ligation (BISIAL) for the synthesis of membrane-associated D-proteins. The BISIAL method allows smooth ligation under strongly chaotropic conditions and the removal of ligation scars on the products, enabling the synthesis of challenging Dprotein targets.

Summary: A method for the preparation of head-to-tail macrocyclic peptides using a dual-intein approach has been reported. The synthetic and genetically encoded fragments are ligated by two native peptide bonds. The study identified a suitable pair of orthogonal inteins and optimized the conditions for a one-pot cyclization protocol, successfully semisynthesizing model macrocyles.

Summary: A new epimerization-free approach for the Fmoc-based synthesis of murepavadin using intramolecular native chemical ligation and desulfurization reaction has been developed, resulting in a high yield of bioactive murepavadin in two steps. The synthetic peptide antibiotic showed potent activity against different clinical isolates of P. aeruginosa. This approach can be easily adapted for the production of murepavadin analogues and other backbone-cyclized peptides.

Summary: Protein splicing is a posttranslational process where an intein removes itself from two exteins. Inteins can be classified as full-length, mini, or split based on their genetic structure. They can also be categorized into three classes according to their splicing mechanisms. Recent evidence suggests that inteins may provide a genetic advantage to host cells by regulating host proteins. Modified inteins have enabled new applications in protein science and synthetic biology by splicing and cleaving their fused exteins.

Summary: 4-Mercaptophenylacetic acid (MPAA) is a commonly used catalyst for native chemical ligation (NCL), but it requires a large excess for practical rates (up to 50-100 equivalents). In this study, we found that the catalytic potency of MPAA can be enhanced by introducing a stretch of arginines in the departing thiol of the thioester. This electrostatically assisted NCL reaction allows for rapid synthesis using substoichiometric concentrations of MPAA, which is advantageous for practical applications.

Summary: Conditional protein splicing is a powerful tool to control the activity of target proteins. In this study, we demonstrated a novel approach using SUMO protease to induce splicing of a split intein. The splicing efficiency was significantly increased by the addition of SUMO protease. This engineered split intein-mediated protein trans-splicing has great potential for controlling protein structure and function in vitro.

Summary: This study reports the development of a novel engineered cyclotide with effective broad-spectrum antibacterial activity against several ESKAPE bacterial strains and clinical isolates. The most active antibacterial cyclotide was extremely stable in serum, showed little hemolytic activity, and provided protection in vivo in a murine model of P. aeruginosa peritonitis. These results highlight the potential of the cyclotide scaffold for the development of novel antimicrobial therapeutic leads for the treatment of bacteremia.
