Inhibition of ferroptosis delays SMC phenotype switch and arterial remodeling, while ferroptosis stress directly triggers SMC dedifferentiation.

Apigetrin, a phytonutrient found in fruits and vegetables, has antioxidant and anti-inflammatory properties. Summary: This study develops an experimental platform for tracking cancer stem cells in colorectal cancer and identifies dormant LGR5(+) cancer stem cells. Transcriptome analysis reveals an upregulation of cell-adhesion molecule COL17A1 in dormant cancer stem cells, and chemotherapy disrupts this dormancy through FAK-YAP activation. Inhibition of YAP signaling prevents chemoresistant cells from exiting dormancy and delays tumor regrowth.

Summary: In this study, a new small molecule library based on 3-methoxy pyrrole was synthesized and one small molecule that can localize in mitochondria and cause damage was identified through screening in colon cancer cells. This small molecule induced mitochondrial damage by generating reactive oxygen species, inhibiting anti-apoptotic protein Bcl-2 and inducing apoptosis and necrosis, leading to significant inhibition of colon cancer cell proliferation while showing minimal toxicity to normal cells. This finding provides a new direction for mitochondria-targeted colon cancer therapy research.

Summary: Non-canonical programmed cell death (PCD) plays an important role in colon cancer, influencing its tumorigenesis, treatment, and prognosis. This review examines the mechanisms, applications, and prospects of different types of non-canonical PCD in colon cancer. Various types of non-canonical PCD, including mitotic catastrophe, ferroptosis, pyroptosis, necroptosis, parthanatos, oxeiptosis, NETosis, PANoptosis, and entosis, are involved in the initiation and development of colon cancer. The review summarizes the association of these non-canonical PCD types with tumorigenesis, progression, prevention, treatments, and prognosis of colon cancer, as well as the potential of drug-resistant colon cancer therapy related to non-canonical PCD and the interaction between different types of non-canonical PCD.
