DLX4 Upregulates TWIST and Enhances Tumor Migration, Invasion and Metastasis

The distal-less homeobox gene 4 (DLX4) is a member of the DLX family of homeobox genes. Although absent from most normal adult tissues, DLX4 is widely expressed in leukemia, lung, breast, ovarian and prostate cancers. However the molecular targets, mechanisms and pathways that mediate the role of DLX4 in tumor metastasis are poorly understood. In this study, we found that DLX4 induces cancer cells to undergo epithelial to mesenchymal transition (EMT) through TWIST. Overexpression of DLX4 increased expression of TWIST expression in cancer cell lines, resulting in increased migratory and invasive capacity. Likewise, knocking down expression of DLX4 decreased TWIST expression and the migration ability of cancer cell lines. DLX4 bound to regulatory regions of the TWIST gene. Both western blotting and immunohistochemistry staining showed that the expression of DLX4 and TWIST are correlated in most of breast tumors. Taken together, these data from both cell models and tumor tissues demonstrate that DLX4 not only upregulates TWIST expression but also induces EMT and tumor metastasis. Altogether, we propose a new pathway in which DLX4 drives expression of TWIST to promote EMT, cancer migration, invasion and metastasis.

LPS/TLR4 Pathways in Breast Cancer: Insights into Cell Signalling

Summary: This review article focuses on the role of toll-like receptors (TLRs), particularly TLR4, in breast cancer. TLRs are widely investigated pattern recognition receptors and TLR4 is the most studied TLR, associated with the development of various cancers, including breast cancer. Activation of TLR4 occurs through the binding of its ligand lipopolysaccharide (LPS), leading to gene expression related to cancer cell proliferation, survival, invasion, and metastasis. The role of TLR4 in breast cancer growth, invasion, and metastasis is attracting significant attention in oncology research, with potential for therapeutic applications.

TQFL12, a novel synthetic derivative of TQ, inhibits triple-negative breast cancer metastasis and invasion through activating AMPK/ACC pathway

PIK3CA hotspot mutations p. H1047R and p. H1047L sensitize breast cancer cells to thymoquinone treatment by regulating the PI3K/Akt1 pathway

Summary: This study evaluated the antitumor effect of thymoquinone, an active component in Nigella sativa, on two PIK3CA hotspot mutations in breast cancer cells and investigated the mechanistic effects of these mutations on the PI3K/Akt1 pathway using molecular dynamics. The findings demonstrated that the mutations reduced the inhibitory effect of Delta Np63 alpha on PIK3CA, leading to enhanced activity of the PI3K downstream signals.

Thymoquinone upregulates IL17RD in controlling the growth and metastasis of triple negative breast cancer cells in vitro

Summary: This study identified IL17RD as a novel target of Thymoquinone (TQ) in triple negative breast cancer (TNBC) cells. TQ treatment inhibited the growth and migration of TNBC cells, and IL17RD overexpression in combination with TQ treatment showed the most effective anti-TNBC effects. Higher expression of IL17RD was associated with longer survival in TNBC patients.

Pharmacological analysis indicated that ADAM17 might be a target for anti-COVID-19 therapy with these compounds.

This review discusses the research progress, application status, and the value of ncRNAs in forensic practice.

Genetic Polymorphism and Population Genetic Structure Analysis of 21 Autosomal STR Loci for a Han-Chinese Population from Luzhou of Southwest China

Summary: This study evaluated 21 autosomal STR loci in 1959 Han-Chinese individuals from Luzhou area and found no substantial linkage disequilibrium among the markers. These loci were in Hardy-Weinberg equilibrium and had high discrimination power and exclusion probability. The results suggest that historical influences, geographical distribution, cultural integration, and economic development may impact the Luzhou Han population and other Chinese populations.

Genetic Polymorphism Analysis of 24 Y-STRs in a Han Chinese Population in Luzhou, Southwest China

Summary: This study analyzed the Y-STRs genetic data of male individuals from the Han population in Luzhou, demonstrating high polymorphism. Comparisons with 11 other related Han populations revealed close genetic relationships between Luzhou Han people and Han populations in Sichuan, Guangdong, and Hunan, consistent with geographical distribution, historical influence, and economic development. These findings are of great value for forensic applications and population genetics research.
