Summary: This study aimed to define the TDP-43/pTDP-43 pathology and evaluate its potential as a tissue biomarker for amyotrophic lateral sclerosis (ALS) and non-ALS cases, by using diagnostic motor nerve biopsies from patients with lower motor neuron syndrome. The researchers found that pTDP-43 aggregates were present in the peripheral nervous system of ALS patients, even in cases with normal features at standard histopathological analysis, suggesting that pTDP-43 aggregation is an early event that may contribute to ALS pathogenesis.

Summary: The Central Nervous System (CNS) is a vital part of the nervous system that comprises the brain and spinal cord. Researchers have proposed a modified version of a neuronal model using a quadratic-based modeling called Digital Spiking Silicon Neuron (DSSN), which can imitate the basic behaviors of the original model. The hardware implementation of the new model shows high similarity with the original model in terms of neuronal behaviors, as well as achieving higher frequency and lower cost conditions.

Summary: 3D cell printing technology is widely studied for medical applications, with a particular focus on neuromuscular tissue. The use of CNSdECM bioink for 3D neuromuscular tissue fabrication shows promising results in supporting motor neuron activity and axon growth. This bioink holds great potential for producing tissue-engineered motor systems using 3D cell printing.

Summary: In insects, thermoreceptors and neurons play critical roles in mediating temperature sensation and thermoresponsive motor behavior. Experimental findings in Drosophila indicate that different functions of ellipsoid body ring neurons R1-R4 are involved in integrating thermosensory information and regulating adaptive motor responses to changes in external temperature. Further research on the neural mechanisms underlying sensory-motor transformation of temperature changes in fruit flies sheds light on the complexity of thermoregulation in ectothermal animals.

Summary: This study found significant differences in blink reflex parameters between ALS patients and healthy controls, suggesting that blink reflex examination could be a promising tool for monitoring disease progression and serving as a prognostic biomarker in ALS patients. Further studies are needed to confirm these findings.

Summary: MicroRNA (miRNA)-mediated gene silencing is a promising method of RNA interference for treating central nervous system diseases. Although several miRNA-based therapeutics have entered clinical trials for various diseases, clinical trials specifically targeting central nervous system diseases are scarce, and effective treatments for these diseases are lacking. This review provides an overview of miRNA-based therapeutics for central nervous system diseases, including their principles, progress, and potential obstacles in drug development, as well as possible solutions for the future.

Summary: Neurons in the central thalamus were found to exhibit different types of responses to repetitive stimuli, including sensory adaptation, increasing suppressive response, and switching activity. Switch-type neurons showed a correlation between their activity before stimulus omission and behavioral latency, suggesting a more advanced role in detecting stimulus omission. These findings imply that the central thalamus may play a crucial role in transmitting integrated signals for temporal information processing in predicting rhythmic event timing accurately.

Summary: Since the discovery of the mechanosensitive Piezo1 channel in 2010, research has been conducted to explore its role in various organ systems. Recent evidence links the activity of Piezo1 channel to the central nervous system. This review systematically summarizes current research on the channel, its implications for central nervous system mechanobiology, and its involvement in disorders.

Summary: The delivery and elimination pathways of intranasally administered nanodrugs were investigated using a PEGylated Fe3O4 nanoparticle labeled with a fluorescent dye. The distribution and elimination of the nanoparticles in the brain were studied through magnetic resonance imaging and fluorescence imaging, providing important insights for therapeutic applications of CNS drugs.

Summary: Spinal muscular atrophy (SMA) is a neuromuscular disease caused by loss of the SMN1 gene and low SMN protein levels. Current gene therapy and clinical trials for SMA have shown significant improvement in the clinical phenotype but not a complete cure. This study demonstrates the independent contribution of peripheral organs to SMA pathology and suggests that treatments should not be restricted to motor neurons.

Summary: This study investigated the potential of liposomes as an efficient drug-delivery system for treating disorders of the central nervous system. By optimizing the liposomes and conducting experiments in a rat model, the study confirmed successful retrograde transport to the spinal cord and dorsal root ganglia. Modifying the liposomes with different substances improved their transport efficiency, providing valuable insights for the future development of novel DDS targeting the CNS.
