Summary: Interferons, IFN-stimulated genes, and inflammatory cytokines are involved in innate immune responses. Knocking-down or knocking-out the expression of IFN alpha-inducible protein 6 (IFI6) enhances the expression of IFNs, ISGs, and pro-inflammatory cytokines after viral infections or poly(I:C) transfection. Overexpression of IFI6 has the opposite effect. IFI6 interacts with RIG-I, affecting RIG-I activation and negatively regulating innate immunity. Targeting IFI6 may be beneficial in treating diseases with excessive innate immune responses and combating viral infections.

Summary: The lack of in vivo models has hindered our understanding of hepatocellular carcinoma (HCC) development. In this study, we used genetic screening in an immunodeficient mouse model to identify oncogenes highly expressed in induced HCC (iHCC) samples. Overexpression of these genes transformed primary human hepatocytes into iHCC. The iHCC model recapitulates histological architecture and gene expression characteristics of clinical HCC samples, providing a biological basis for studying HCC initiation and identifying diagnostic biomarkers and targets for cellular immunotherapy.

Summary: There is increasing evidence showing the crucial role of the immune microenvironment, particularly the polarization state and function of macrophages, in the development of hepatocellular carcinoma. Tumor-derived exosomes in hepatocellular carcinoma act as information carriers and regulate the physiological state of cells in the microenvironment, thus controlling cancer progression. This review focuses on the role of exosome content in disease outcomes at different stages of hepatitis B virus/hepatitis C virus-induced hepatocellular carcinoma, and explores the mechanism by which macrophages contribute to hepatocellular carcinoma formation and the regulation of macrophage functions by the heterogeneity of exosome loading. The application prospects of exosome-based targeted drug delivery in immunotherapy research on hepatocellular carcinoma are also summarized.

Summary: In this study, a naphthyridine-based molecule, CMBL4c, was found to bind to the RNA structure formed by CGG repeats in FXTAS cells, reducing the synthesis of FMRpolyG and nuclear inclusions in affected cells. Additionally, delivery of CMBL4c also decreased FMRpolyG-mediated cytotoxicity and apoptosis.
