Intersectin 1 (ITSN1) is a multidomain adaptor protein that functions in clathrin-mediated endocytosis and signal transduction. This protein is highly abundant in neurons and is implicated in Down syndrome, Alzheimer's disease and, possibly, other neurodegenerative disorders. Here we used an in vitro binding assay combined with MALDI-TOF mass spectrometry to identify novel binding partners of ITSN1. We found that the neuron-specific isoform of the stable tubule-only polypeptide (STOP) interacts with SH3A domain of ITSN1. STOP and ITSN1 were shown to form a complex in vivo and to partially co-localize in rat primary hippocampal neurons. As STOP is a microtubule-stabilizing protein that is required for several forms of synaptic plasticity in the hippocampus, identification of this interaction raises the possibility of ITSN1 participation in this process. The atomic-resolution crystal structure of the oligomerization region peptide revealed an antiparallel coiled-coil interface supporting NT-NT domain interactions in Arc oligomerization. The study also showed that Arc mRNA enhances higher-order oligomerization dependent on the oligomerization motif, leading to the formation of virus-like capsids.

Summary: We have demonstrated the expression of NMDAR during the early stages of cancer, specifically HCC, in the cell transformation process. The aim of our study was to define the disease stages from precancerous liver disease to liver cancer progression when NMDAR subunits were expressed/detected. A fibrosis/HCC rat model, immunohistochemistry combined with epifluorescence microscopy imaging, and Western blotting were employed. The dynamics of appearance of NMDAR subunits, their expression, and methylation status during the development of HCC were shown and discussed.

The molecular characteristics of high-grade gastroenteropancreatic neuroendocrine neoplasms

Summary: High-grade gastroenteropancreatic neuroendocrine neoplasms (GEP-NEN) are a rare and understudied type of tumors, with a very poor prognosis. Molecular analysis of 181 HG GEP-NEN patients revealed unique genetic alterations and copy number changes in different subgroups of tumors, indicating potential personalized treatment options. Limited similarities were found with small-cell lung cancer (SCLC), suggesting the need for subtype-specific therapeutic approaches.

Summary: The most common inherited cause of ALS and FTD is the expansion of GGGGCC repeats in the C9orf72 gene. In addition to haploinsufficiency and toxic RNA foci, other disease mechanisms are involved. Through the use of the BioID approach, it was found that DPR aggregates are associated with specific proteins that play important roles in protein folding and degradation. Heat shock proteins and co-chaperones were found to be closely associated with one of the DPRs, poly-GA, and may play a significant role in reducing its aggregation.

Ca2+-Permeable AMPA Receptors Contribute to Changed Dorsal Horn Neuronal Firing and Inflammatory Pain

Summary: The dorsal horn (DH) neurons of the spinal cord play a critical role in processing nociceptive input in the central nervous system. The hyperexcitability of DH neurons has been linked to Ca2+-permeable AMPA receptors (AMPARs) and contributes to nociceptive hypersensitivity. Inflammatory pain conditions result in changes in action potential (AP) parameters in DH neurons, and selective inhibition of Ca2+-permeable AMPARs can alleviate nociceptive hypersensitivity.

Summary: The study found that members of the nuclear import receptor protein family can reduce the formation of pathological TDP-43 aggregates. Using KPNB1 as a model, it was discovered that its activity depends on the prion-like C-terminal region of TDP-43, mediating co-aggregation with nucleoporins such as Nup62. KPNB1 acts as a molecular chaperone in these co-aggregates, reversing the aberrant phase transition of Nup62 and TDP-43.

Elucidating afferent-driven presynaptic inhibition of primary afferent input to spinal laminae I and X

Summary: Although our understanding of how afferent-driven presynaptic inhibition shapes peripheral input to nociceptive neurons is limited, this study examined this effect on primary afferent input to spinal neurons in two nociceptive-processing regions. The results showed that attenuation of presynaptic inhibition resulted in the appearance of new mono- and polysynaptic excitatory postsynaptic current components. These mechanisms are important for shaping primary afferent input to neurons in the spinal nociceptive-processing network.

Treatment of Acute Respiratory Distress Syndrome Caused by COVID-19 with Human Umbilical Cord Mesenchymal Stem Cells

Summary: This study investigated the effects of mesenchymal stem cell (MSC) transplantation on severe COVID-19 patients. The results showed that triple MSC transplantation was safe and did not cause severe adverse reactions. CT imaging revealed that MSC transplantation resulted in significantly lower fibrotic changes in the lung compared to conventional anti-viral treatment. Additionally, MSC therapy improved lymphocyte recovery and reduced inflammation markers in the peripheral blood.

5-Deazaalloxazine as photosensitizer of singlet oxygen and potential redox-sensitive agent

Summary: This article reports the photophysical properties of 5-deazaalloxazine (1a) in different solvents. It is found that the triplet excited states of 1a in water and acetonitrile have microsecond lifetimes, and 1a can efficiently generate singlet oxygen under oxygen conditions. In addition, experiments using human red blood cell models show that 1a has antioxidant and cytotoxic activities.

Comparative alternative polyadenylation profiles in differentiated adipocytes of subcutaneous and intramuscular fat tissue in cattle

Genome-wide regulation of Pol II, FACT, and Spt6 occupancies by RSC in Saccharomyces cerevisiae

Summary: RSC is an essential ATP-dependent chromatin remodeling complex in Saccharomyces cerevisiae. It regulates nucleosome-depleted regions (NDRs) by sliding flanking nucleosomes away from NDRs. Depletion of RSC leads to nucleosome encroachment in NDRs and transcription initiation defects. The study compared the effects of catalytic-dead Sth1 and rapid depletion of Sth1 on transcription. Rapid depletion of Sth1 reduces recruitment of TBP and Pol II, while the catalytic-dead mutant exhibits a severe reduction in TBP binding but accumulates Pol II in coding regions. The results suggest a role for RSC in transcription elongation and termination processes.

Mapping the genetic architecture of idiopathic pulmonary fibrosis: Meta-analysis and epidemiological evidence of case-control studies

Age-different BMSCs-derived exosomes accelerate tendon-bone interface healing in rotator cuff tears model
