Summary: Immunoprofiling for identifying biomarkers and integrating with clinical trial outcomes play a critical role in improving immunotherapy for cancer patients. The CIMAC-CIDC Network aims to establish an infrastructure for systematic biomarker identification and correlation with clinical outcomes across trials, facilitated by harmonized assays and computational expertise. By establishing this network, immuno-oncology biomarker development and cross-trial data analysis can be enhanced.

Inflamed Ulcerative Colitis Regions Associated With MRGPRX2-Mediated Mast Cell Degranulation and Cell Activation Modules, Defining a New Therapeutic Target

Philip Schumm, Talin Haritunians, Dermot P. McGovern, Yuval Itan, Judy H. Cho

Summary: The research reveals that in human UC, inflamed regions are distinguished by MRGPRX2-mediated activation of mast cells, with decreased activation observed with a UC-protective genetic variant. These results provide important clues for the cell modules of UC activation and a new therapeutic target.

Food colorants metabolized by commensal bacteria promote colitis in mice with dysregulated expression of interleukin-23

Summary: The study demonstrates that azo dyes Red 40 and Yellow 6 can induce IBD-like colitis in mice expressing IL-23, with the microbiota playing a role in metabolizing the dyes into harmful substances.

MMP2 and TLRs modulate immune responses in the tumor microenvironment

Luciana R. Muniz-Bongers, Christopher B. McClain, Mansi Saxena, Gerold Bongers, Miriam Merad, Nina Bhardwaj

Summary: The study revealed that MMP2 plays a promoting role in melanoma tumor growth by reducing cytolytic cells and increasing M2 macrophages. On the other hand, knockdown of MMP2 slows tumor growth and enhances T cell proliferation and NK cell recruitment. These effects of MMP2 are mediated through dysfunctional DC-T cell cross-talk.

Precise quantification of bacterial strains after fecal microbiota transplantation delineates long-term engraftment and explains outcomes

Varun Aggarwala, Ilaria Mogno, Zhihua Li, Chao Yang, Graham J. Britton, Alice Chen-Liaw, Josephine Mitcham, Gerold Bongers, Dirk Gevers, Jose C. Clemente, Jean-Frederic Colombel, Ari Grinspan, Jeremiah Faith

Summary: Researchers developed a method named Strainer to detect and track bacterial strains in recipients using a large collection of bacterial strains and metagenomic sequencing data, finding that 71% of donor microbiota strains stably engrafted in recipients up to 5 years post-FMT and significantly influenced clinical outcomes.

Circulating bioactive bacterial DNA is associated with immune activation and complications in common variable immunodeficiency

Hsi-en Ho, Lin Radigan, Gerold Bongers, Ahmed El-Shamy, Charlotte Cunningham-Rundles

Summary: Circulating bacterial 16S rDNA belonging to gut commensals is significantly increased in CVID serum, especially in patients with inflammatory manifestations. Levels of serum bacterial DNA are associated with parameters of systemic immune activation, increased serum IFN-gamma, and the lowest numbers of isotype-switched memory B cells, potentially leading to chronic immune activation.
