Summary: This study identifies a membrane protein, PODXL, as a critical regulator of pluripotency in stem cells. It is found that PODXL plays a role in regulating cholesterol levels, which in turn affects the function and characteristics of pluripotent stem cells. Additionally, PODXL is involved in modulating actin network, SREBP transcription factors, and lipid raft dynamics.

Summary: The glycoprotein mesothelin is overexpressed in ovarian cancer and may play a role in tumor progression by affecting the peritoneal ultrastructure and mesothelial cell surface architecture. Reduced expression of host mesothelin could result in smaller tumor aggregates and decreased peritoneal metastatic tumor burden in mice.

Summary: Researchers have developed an antibody-based T-cell engager that can target mesothelin and discriminate high mesothelin-expressing tumors from normal tissue. This engager is also effective in the presence of soluble mesothelin. When used in combination with a checkpoint inhibitor, it shows enhanced tumor suppression and T-cell infiltration. This new approach may overcome the limitations of previous mesothelin-targeted immuno-oncology drugs.

Summary: The T cell receptor fusion construct gavocabtagene autoleucel shows significant anti-tumor activity in patients with treatment-refractory mesothelin-expressing solid tumors. However, it may have a narrow therapeutic window.

Summary: This article reviews the clinical research status, obstacles, advancements, and challenges of anti-MSLN CAR-T cell therapy, and summarizes strategies to improve its efficacy and safety. Anti-MSLN CAR-T cell therapy is a rapidly developing immunotherapy that has shown high safety but limited efficacy in clinical trials. Current approaches to enhance the proliferation and persistence of anti-MSLN CAR-T cells include local administration and the introduction of new modifications.

Summary: Stable Sf9 cell pools were developed for producing HIV-1 Gag-eGFP virus-like particles, which showed a significantly increased fluorescence intensity and VLP production. The metabolism analysis indicated no significant differences between stable cell pools and parental Sf9 cells, highlighting the flexibility of these cells in adapting to different culture conditions for recombinant protein production.

Summary: Our study demonstrates the role of PspA in bacterial membrane remodeling and vesicular structure formation through cryo-EM analysis. The findings suggest that PspA belongs to the evolutionary ancestry of ESCRT-III proteins involved in membrane fusion and fission activities. This sheds light on the functional properties of bacterial PspA/IM30/LiaH proteins.

Summary: This study introduces a reporter-guided metabolic engineering tool based on random mutagenesis, selective pressure, and single-cell sorting for actinomycetes. It enables the identification of high-performing individual cells from mutant populations and has been successfully applied to strain development and drug discovery. The method allows for the activation of silent metabolic pathways and increased yields of proteins and natural products.

Summary: BPFP1 is a new tool for Bcl-2 protein quantification, carcinoma cell visualization, and cell sorting. It can fluorescently label carcinoma cells by binding to overexpressed Bcl-2 protein and distinguish carcinoma cells from normal tissue cells based on the different Bcl-2 protein expression levels.
