The optokinetic response in zebrafish and its applications

The optokinetic response (OKR) is a stereotyped eye movement in response to movement in he surround. The OKR serves to stabilize the visual image on the retina, and allows for high resolution vision. Due to its high selection value, all vertebrates display this basic behavior. Here, we review the properties of the OKR with a focus on the zebrafish, including methodological aspects of measuring eye movements in small larvae. The genetic amenabilities of the zebrafish model permit the use of this reflexive behavior in genetic screens. Such approaches have led to the isolation of mutant strains with specific defects in the visual pathway. In addition to the use of the OKR as a screening assay, mutations with characteristic abnormalities in the execution of this behavior will enable the analysis of sensory-motor control in great detail. A case in point is the belladonna mutation, where an axonal misrouting effect at the optic chiasm leads to a reversed OKR with a number of interesting properties.

Summary: The loss of function in the astroglia predominant eaat2a gene in zebrafish leads to altered neuronal and astroglial networks, resulting in epileptic seizures and increased extracellular glutamate levels. Interestingly, while hyperexcitability is observed during seizures, basal neuronal and astroglial activity is reduced, leading to decreased overall locomotion in eaat2a(-/-) mutant animals.

Hyper-reoriented walking in minimal space

Summary: Hyper-reoriented walking is a new reorientation technique that reduces the physical space required in VR applications by allowing users to walk along gridlines with twice the speed of real walking and perform turns at cross-points with half the rotation speed in physical space. Experiment results showed that walking accuracy was good, but a correction algorithm was needed for slight errors when turning at T-junctions in a grid-like labyrinth.

Summary: This study identified potential targets of manganese neurotoxicity using a slc39a14(-/-) mutant zebrafish model exposed to MnCl2. The findings suggest that dysregulation of Ca2+ homeostasis and activation of the unfolded protein response are key features of manganese neurotoxicity. Additionally, manganese exposure led to locomotor defects and changes in visual and neuronal activity.

Summary: Imaging of large, cleared samples is achieved using a multi-immersion 'Schmidt objective' consisting of a spherical mirror and an aspherical correction plate. This objective is compatible with all homogeneous immersion media and achieves an NA of 1.08 at a refractive index of 1.56, with a 1.1-mm FOV and 11-mm WD.

SonoRotor: An Acoustic Rotational Robotic Platform for Zebrafish Embryos and Larvae

Summary: This study presents a miniaturized acoustic rotation platform, called SonoRotor, for animal models. By controlling the voltage applied to the device, the rotation speed of zebrafish can be adjusted. The developed acoustic rotation manipulation platform is simple, cost-effective, allows multiview imaging, high-throughput operation, and can be combined with downstream analytical techniques to perform various biological micromanipulations on small animal models.

Neutrophil extracellular trap formation during surgical procedures: a pilot study

Summary: In this study, the impact of surgical trauma on NETosis and monocytic HLA-DR expression was investigated. The results showed that NETosis decreased after surgery, with a more pronounced decrease in ACB patients. ACB surgery also led to a down regulation of monocytic HLA-DR expression. These findings suggest that the ability of neutrophils to form NETs may be compromised following severe surgical trauma, and there may be a post-operative immunosuppressed state.
