Ceruloplasmin and myeloperoxidase in complex affect the enzymatic properties of each other

Ceruloplasmin (CP), the multicopper oxidase of plasma, interacts with myeloperoxidase (MPO), an enzyme of leukocytes, and inhibits its peroxidase and chlorinating activity. Studies on the enzymatic properties shows that CP behaves as a competitive inhibitor impeding the binding of aromatic substrates to the active centre of MPO. The contact between CP and MPO probably entails conformational changes close to the p-phenylenediamine binding site in CP, which explains the observed activation by MPO of the substrate's oxidation. CP subjected to partial proteolysis was virtually unable to inhibit activity of MPO. The possible protein protein interface is comprised of the area near active site of MPO and the loop linking domains 5 and 6 in Cl One of the outcomes of this study is the finding of a new link between antioxidant properties of CP and its susceptibility to proteolysis.

Fluorescent Probes for HOCl Detection in Living Cells

Summary: Hypochlorous acid (HOCl) plays a crucial role in the immune system but its high reactivity can also lead to the development of diseases. Therefore, developing highly sensitive and selective HOCl biosensors is important. This review summarizes the achievements in the development of chemosensors for HOCl detection in biological systems from 2016 to 2021. The challenges in designing and applying small-molecule probes are discussed, and recommendations are given for their future application.

Spike protein competes with transferrin receptor 1 (TfR1) ligands and shows cross-reactivity with lactoferrin (Lf) and transferrin (Tf). Apo-Lf and holo-Lf can inhibit SARS-CoV-2 replication and protect against COVID-19-induced pathogenesis.

Summary: Hyperglycemia in diabetes mellitus alters the functionality of neutrophils, reducing their bactericidal activity mediated by MPO, which is affected by the modification of proteins by glucose and its derivative methylglyoxal. This phenomenon may contribute to impaired wound healing and increased susceptibility to infection in patients with hyperglycemia.

Summary: This study investigated the effect of incorporating pectin into vaterite microparticles on cytotoxicity and mucin-dependent neutrophil activation. The results showed that the inclusion of pectin did not cause damage to neutrophils and did not have direct pro- or antioxidant effects. However, when neutrophils were activated by mucin-treated microparticles, the incorporation of pectin into vaterite microspheres enabled an antioxidant effect to be reached, presumably via exposed ligands.

Summary: In this study, the impact and mechanism of streptococcal arginine deiminase (ADI) on the function of human peripheral blood T lymphocytes were investigated. The results showed that ADI decreased arginine concentration, suppressed immune cell activity and altered cell differentiation state, as well as inhibited autophagy processes.

X-Ray Analysis of the Monoclinic Crystal Form of Human Ceruloplasmin

Summary: This study successfully obtained human ceruloplasmin crystals and determined the conformation of a labile functional loop between two structural domains. By comparing different crystal packings, the efficiency of the protein purification method was confirmed, along with the importance of loop integrity in neutrophil myeloperoxidase inhibition during complex formation with ceruloplasmin.
