Oxidative stresses induced by glycoxidized human or bovine serum albumin on human monocytes

Oxidative stress and protein modifications are frequently observed in numerous disease states. Albumin, the major circulating protein in blood, can undergo increased glycoxidation in diabetes. Protein glycoxidation can lead to the formation of advanced glycoxidation end products, which induce various deleterious effects on cells. Herein, we report the effect of glucose or methylglyoxal-induced oxidative modifications on BSA or HSA protein structures and on THP1 monocyte physiology. The occurrence of oxidative modifications was found to be enhanced in glycoxidized BSA and HSA, after determination of their free thiol group content, relative electrophoretic migration, carbonyl content, and antioxidant activities. Cells treated with glycoxidized albumin exhibited an overgeneration of intracellular reactive oxygen species, impairments in proteasomal activities, enhancements in PAGE expression, and an accumulation of carbonylated proteins. These novel observations made in the presence of a range of modified BSA and HSA facilitate the comparison of the glycoxidation extent of albumin with the oxidative stress induced in cultured monocytes. Finally, this study reconfirms the influence of experimental conditions in which AGEs are generated and the concentration levels in experiments designed to mimic pathological conditions. Summary: This study evaluated the antioxidant and cytoprotective effects of polyphenol-rich extracts from two medicinal plants, A. borbonica and D. apetalum, from Reunion Island. The results showed that the polyphenol extracts of these plants could limit LDL oxidation and intracellular oxidative stress, and protect the endothelium.

Summary: This study evaluated the effects of polyphenols extracted from Antirhea borbonica on obesity and diabetes-related cerebrovascular damages during stroke. The results showed that polyphenols reduced insulin resistance, hyperglycemia, and dyslipidemia caused by obesity, and attenuated brain infarct, hemorrhagic transformation, and blood-brain barrier disruption. Polyphenols also exhibited anti-inflammatory and antioxidant properties, and decreased MMP-2 activity in hyperglycemic condition. Furthermore, the protective effects of polyphenols were associated with their circulating metabolites.

Summary: This study aimed to improve the bioavailability of curcumin and evaluate the synergistic effect of curcumin-reconstituted high-density lipoprotein nanoparticles on MGO-induced endothelial dysfunction in diabetic patients. The results showed that Cur-rHDL nanoparticles can alleviate MGO-induced cytotoxicity and oxidative stress, protect endothelial cell barrier integrity, and enhance the bioavailability of curcumin.

Biochemical markers for low glycemic index and approaches to alter starch digestibility in rice

Summary: Recent studies suggest that dietary fibre rich wholegrain foods with low starch digestibility and high resistant starch content may contribute to low glycemic index and glycemic load. To prevent the development of type-II diabetes, rice based low GI foods with high resistant starch content have been developed. The increase in amylose and resistant starch content in rice, along with the action of starch debranching enzyme and the inhibition of starch digestibility by phytic acid, have been achieved through mechanical processing and modern technologies like CRISPR.
