Summary: The study found that trehalose dried from alcohol as a solvent has lower crystallization temperature and specific crystallization characteristics compared to those dried from water. Summary: In this study, a method was developed to predict wg' using the thermodynamic model PC-SAFT and one experimental data point of Tg' for (1) single excipients, (2) given compositions of a binary excipient mixture, and (3) single excipients in aqueous (model) protein solutions. The results show that this new approach can accurately predict wg' in the systems considered, including the non-linear course of wg' identified for different sucrose/ectoine ratios, as well as the course of wg' as function of the protein concentration. This newly developed method greatly reduces the experimental effort required.

Summary: Nanocrystal dispersions have been found to improve the bioavailability of the deuterated pyrazoloquinolinone ligand DK-I-56-1. The freeze-drying process using different combinations of lyoprotectant and bulking agent was investigated to improve the storage stability of the formulation. The results showed that the appropriate combination of sucrose or trehalose with mannitol, along with the selection of lyophilization process parameters, could yield nanocrystals with satisfactory stability.

Summary: Amino acids-based co-amorphous system (CAM) is a promising approach to overcome the dissolution challenge of biopharmaceutics classification system class II drugs. However, the understanding of non-ionic based molecular interactions is still limited, and there is a lack of studies on the drug-to-co-former ratio and the development of scalable manufacturing processes. Freeze-drying using the tert-butyl alcohol-water cosolvent system has shown to produce an optimal non-salt-based class II drug-amino acid CAM.

Sucrose was found to be a more effective stabilizer against freeze-thaw stress than mannitol, demonstrating the importance of an amorphous matrix. NaCl presence reduced the aggregation propensity of nanoparticles, while the freezing step also impacted aggregation. Surfactants did not necessarily improve colloidal stability and could limit storage stability due to phase separation, but a specific cryoprotectant mixture could overcome this hurdle.

Summary: In this study, a biocomposite made of alginate and gelatin crosslinked with genipin was developed for immobilizing beta-galactosidase. The microstructure, thermal stability, and operational stability of the immobilized enzyme were analyzed using thermal analyses and SAXS. The results showed that the addition of genipin and enzyme significantly reduced the size and density of the Ca(II)-alginate rods in the biocomposite. The immobilized beta-galactosidase exhibited high storage stability and retained a high relative activity after multiple reuses in lactose hydrolysis.

Summary: The influence of organic and inorganic anions on the Maillard reaction in glucose-glycine systems was studied under conditions of 100 degrees C and pH close to neutrality. The progress of the Maillard reaction was measured using spectrophotometric, pH changes, and colorimetric methods. Different trends were observed depending on the nature of the anion. Anions with basic properties showed an exponential correlation with the Maillard reaction kinetic constants, while anions with no acid-base properties exhibited a linear correlation with the water dissolution enthalpies of their sodium salts.
