Trehalose-water-salt interactions related to the stability of β-galactosidase in supercooled media

The conservation of desirable properties in foods and ingredients is often based on the maintenance of the amorphous metastable properties of the systems. Enzymes may be stabilized by drying in saccharide matrices, but a second excipient is generally required to improve sugar protective effects. The effect of electrolytes on the thermophysical properties of sugar systems is of special interest because of their major influence on water structure and their possible interactions with biomolecules. Salts affect the kinetics of very important changes in sugar systems such as crystallization. The purpose of the present work was to analyze fungal beta-galactosidase stability in supercooled systems of trehalose-containing electrolytes (water soluble acetates, citrates, and chlorides of magnesium and potassium). The degree of sugar crystallization was also related to enzyme stability. Potassium citrate and acetate improved enzyme stability during freeze-drying and thermal treatment of samples at water activity (a(w)) of 0.22. However, trehalose crystallization inhibition at a(w)=0.43 (which was about 50-60%, related to the system without salt) impaired enzyme protection. Certain salts may act retarding sugar crystallization, but in the presence of salts, trehalose crystallization is even more critical because the enzyme is confined in a highly salt-concentrated region. Thus, crystallization inhibition by sugar-salt combinations should be carefully conducted.

Development of a biocomposite based on alginate/gelatin crosslinked with genipin for β-galactosidase immobilization: Performance and characteristics

Summary: In this study, a biocomposite made of alginate and gelatin crosslinked with genipin was developed for immobilizing beta-galactosidase. The microstructure, thermal stability, and operational stability of the immobilized enzyme were analyzed using thermal analyses and SAXS. The results showed that the addition of genipin and enzyme significantly reduced the size and density of the Ca(II)-alginate rods in the biocomposite. The immobilized beta-galactosidase exhibited high storage stability and retained a high relative activity after multiple reuses in lactose hydrolysis.

Physicochemical transformations of corn-based snacks with underexploited grains addition during storage: multi-analytical assessment

Revaluing Anion Effects on Maillard Reaction Catalysis as a Relevant Feature for Kinetic Predictions

Summary: The influence of organic and inorganic anions on the Maillard reaction in glucose-glycine systems was studied under conditions of 100 degrees C and pH close to neutrality. The progress of the Maillard reaction was measured using spectrophotometric, pH changes, and colorimetric methods. Different trends were observed depending on the nature of the anion. Anions with basic properties showed an exponential correlation with the Maillard reaction kinetic constants, while anions with no acid-base properties exhibited a linear correlation with the water dissolution enthalpies of their sodium salts.

Evaluation of SIMCA and PLS algorithms to detect adulterants in canola oil by FT-IR

Summary: In this study, adulteration of canola oil with four potential edible oils was analyzed using FT-IR and chemometric methods. Excellent classification results were obtained with multi-class approaches, while the selection of variables was necessary to improve the classification error with one-class approaches. The differences observed using different methods were related to the nature of each model depending on how the boundaries are set in each of them.
