Chlorogenic acid (ChA) exhibits a multitude of positive health effects, however, the signaling mechanisms by which ChA could influence the inflammatory response in experimental colitis are unknown. To answer this question, we induced colitis in mice by administration of 2.5% dextran sulfate sodium (DSS) in drinking water for seven days. Oral administration of ChA significantly ameliorated clinical symptoms, improved disease activity index and colon shortening induced by DSS. Furthermore, ChA administration resulted in a suppression of phosphorylated extracellular signal-regulated kinases 1 and 2 (ERK1/2), c-Jun N-terminal kinases 1 and 2 (JNK1/2), Akt and signal transducer and activator of transcription 3 (STAT3) with concomitant upregulation of phosphatase and tensin homolog (PTEN) expression. Immunohistochemical analysis showed a dose-dependent decrease in expression and nuclear translocation of nuclear factor-kappa B (NF-kappa B) p65 subunit, which was accompanied by suppression of pro-inflammatory cytokine tumor necrosis factor-alpha (TNF-alpha) expression. Induction of apoptosis and oxidative stress was attenuated in a dose-dependent manner by suppressing Bax, caspase-8, caspase-9 and heme oxygenase-1 (HO-1) protein expression in mice administrated with ChA. The results of the current study suggest that ChA could be useful for the treatment of inflammation and attenuating colitis severity by suppressing activation of pro-inflammatory and apoptotic signaling pathways.

Profiling Colorectal Cancer in the Landscape Personalized Testing-Advantages of Liquid Biopsy

Donatella Verbanac, Andrea Ceri, Iva Hlapcic, Mehdi Shakibaei, Aranka Brockmueller, Bozo Kruslin, Neven Ljubicic, Neven Barsic, Dijana Detel, Lara Baticic, Lada Rumora, Anita Somborac-Bacura, Mario Stefanovic, Ivana Celap, Alma Demirovic, Roberta Petlevski, Jozsef Petrik, Marija Grdic Rajkovic, Andrea Hulina-Tomaskovic, Ivana Rako, Luciano Saso, Karmela Barisic

Summary: Drug-specific therapeutic approaches for colorectal cancer have significantly improved patient health, but there is a need for personalized treatments based on genetic and epigenetic tumor profiles. Circulating tumor DNA analysis has proven inaccurate, but detecting circulating tumor cells and protein analysis of exosomes show promise. Blood liquid biopsy has potential for the future, if results align with tissue analyses.

Diagnostic accuracy of three SARS-CoV2 antibody detection assays, neutralizing effect and longevity of serum antibodies

Summary: Currently, there is insufficient evidence to determine whether SARS-CoV-2 antibodies provide protection from future infection and the duration of immunity. The kinetics of the immune response to SARS-CoV-2 infection and the role of serology in estimating individual protective immunity have yet to be established.

Gender-Related Differences in BMP Expression and Adult Hippocampal Neurogenesis within Joint-Hippocampal Axis in a Rat Model of Rheumatoid Arthritis

Summary: This study demonstrates gender differences in the expression and effects of immune modulatory molecules BMPs during the inflammatory process of rheumatoid arthritis in an experimental animal model. Male rats exhibited earlier disease onset and recovery but a worse clinical course in the early phases, while female rats showed worsening clinical symptoms during remission. Females exhibited higher expression related to synovial inflammation during remission, with stronger correlations with serum cytokines, hippocampal, and neurogenesis.

Summary: This study found that Oleanolic acid (OA) can induce both apoptosis and prosurvival mitophagy in colon cancer cells through the p38/FOXO3a/Sirt6 signaling pathway. Additionally, OA can chemosensitize colon cancer cells to 5-fluorouracil (5-FU).

Significance of the Lung Immune Prognostic Index for Assessment of the Reliability of the Clinical Treatment Outcome for Advanced Non-Small-Cell Lung Cancer in Patients with COVID-19 Infection

Kristina Krpina, Martina Mavrinac, Miroslav Samarzija, Ena Tolic, Dora Darapi, Lara Baticic

Summary: This study investigates the clinical significance and predictive role of the Lung Immune Prognostic Index (LIPI) in advanced non-small cell lung cancer (NSCLC) patients eligible for immunotherapy in combination with chemotherapy. It also examines the association between COVID-19 infection and the course and outcome of NSCLC treatment. The results show that LIPI has predictive power and clinical relevance as a biomarker in advanced NSCLC, and patients infected with COVID-19 have a higher survival rate.

Summary: Cardiac surgery is a high-risk procedure that often leads to endothelial injury and subsequent organ dysfunction. This review focuses on the structure and function of the endothelial glycocalyx and shedding mechanisms in cardiac surgery, as well as potential strategies to protect and restore it. Additionally, the latest evidence on biomarkers of endothelial dysfunction and their clinical implications in cardiac surgery patients is summarized.
